血漿長鏈非編碼RNA HIT對乳腺癌的診斷價值

【摘要】:目的探討轉化生長因子-β(TGF-β)誘導的HOXA轉錄物(HOXA transcript induced by TGF-β,HIT)在乳腺癌女性血漿中的表達情況及其對乳腺癌的潛在診斷價值。方法收集乳腺癌女性患者血漿標本124例(乳腺癌組)、乳腺良性疾病患者血漿標本59例(乳腺良性疾病組)及健康體檢女性血漿標本48例(對照組),采用實時定量逆轉錄PCR(real time RT-PCR)方法分別檢測各組標本中HIT的表達水平,分析HIT與乳腺癌臨床病理特征的相關性,繪制受試者工作特征曲線(ROC)以評估血漿HIT對乳腺癌的診斷效能。結果 HIT在乳腺癌組血漿中表達水平[0.12(0.02,0.33)]明顯低于對照組[0.55(0.20,1.59)],差異有統(tǒng)計學意義(U=1 020,P0.000 1),HIT在乳腺良性疾病組血漿中表達水平[0.10(0.02,0.19)]明顯低于對照組,差異有統(tǒng)計學意義(U=308.5,P0.000 1),HIT在乳腺癌組血漿中表達水平低于乳腺良性疾病組,但差異無統(tǒng)計學意義(U=3 028,P=0.060 2)。血漿中HIT的表達水平與雌激素受體(ER)(P=0.021)和淋巴結轉移(P=0.033)明顯相關。血漿HIT診斷乳腺癌的ROC曲線下面積(AUC)為0.799,敏感度和特異度分別為83.3%和66.7%。結論血漿中低表達的lncRNA HIT可能成為乳腺癌診斷的一個潛在的新型生物標志物。
[Abstract]:Objective to investigate the expression of HOXA transcript induced by TGF- 尾 in the plasma of women with breast cancer and its potential diagnostic value. Methods the plasma samples of 124 women with breast cancer were collected. The expression of HIT was detected by real-time quantitative reverse transcription PCR(real time RT-PCR method in 59 plasma samples from patients with adenocarcinoma, 59 patients with benign breast disease (benign breast disease) and 48 healthy women (control group). To analyze the correlation between HIT and clinicopathological features of breast cancer. Results the level of plasma HIT expression in breast cancer group [0.12 0. 02 鹵0. 33] was significantly lower than that in control group [0. 55 0. 20 鹵1. 59], and the difference was statistically significant. The expression level of plasma in the disease group was significantly lower than that in the control group. The difference was statistically significant (P < 0.05). The plasma level of Hit in breast cancer group was lower than that in benign breast disease group. However, there was no significant difference between the two groups. The expression of HIT in plasma was significantly correlated with that of estrogen receptor ERP (0.021) and lymph node metastasis (P < 0.033). The area under the ROC curve of plasma HIT for diagnosis of breast cancer was 0.799, the sensitivity and specificity were 83.3% and 83.3%, respectively. Conclusion the low expression of lncRNA HIT in plasma may be a potential new biomarker for diagnosis of breast cancer.
