【摘要】：目的研究整合素α5(integrinα5,ITGA5)和caspase 3在肝癌組織中的表達及其臨床意義。方法1.免疫組化Elivision二步法檢測ITGA5和caspase 3在36例肝癌組織石蠟標本、23例癌旁組織石蠟標本中的表達。分析ITGA5、caspase 3在肝癌組織中的表達與臨床病理特征的關(guān)系。2.熒光定量RT-PCR、Western blot分別檢測經(jīng)外科手術(shù)切除凍存的25例肝細胞癌患者的癌組織及癌旁組織中ITGA5和caspase 3 mRNA及蛋白質(zhì)的相對表達量。3.SPSS20.0統(tǒng)計軟件用于分析實驗數(shù)據(jù),計量資料以ⅹ_結(jié)果1.免疫組化染色結(jié)果±s表示,兩組間比較采用獨立樣本t檢驗,計數(shù)資料比較采用卡方或Fisher精確概率法,兩變量關(guān)聯(lián)性采用Spearman相關(guān)性分析,以P0.05為差異有統(tǒng)計學意義。1.1 ITGA5蛋白質(zhì)主要表達于細胞膜和細胞質(zhì),其在肝癌組織和癌旁組織中的陽性表達率分別為88.89%(32/36),43.48%(10/23),差異有統(tǒng)計學意義(χ2=14.109,P0.01)。ITGA5在肝癌中的表達與TNM分期及腫瘤轉(zhuǎn)移有關(guān)(P均0.05)。TNM分期為Ⅲ~Ⅳ期的肝癌組織ITGA5陽性表達率更高;伴有轉(zhuǎn)移的肝癌組織ITGA5陽性表達率更高。1.2 caspase 3蛋白質(zhì)主要表達于細胞質(zhì),其在肝癌組織和癌旁組織中的陽性表達率分別為30.56%(11/36),86.96%(20/23),差異有統(tǒng)計學意義(χ2=17.903,P均0.01)。caspase 3在肝癌中的表達與HBsAg、TNM分期及腫瘤轉(zhuǎn)移有關(guān)(P均0.05)。HBsAg陽性、TNM分期為Ⅲ~Ⅳ期、伴有轉(zhuǎn)移的肝癌組織caspase 3陽性表達率更低。2.熒光定量RT-PCR檢測結(jié)果顯示:ITGA5 mRNA在肝癌組織中的相對表達量(3.188±1.339)高于癌旁組織中的相對表達量(0.388±0.238);caspase 3 mRNA在肝癌組織中的相對表達量(0.209±0.108)低于癌旁組織中的相對表達量(5.755±2.522)。差異均有統(tǒng)計學意義(P均0.01)。3.Western blot檢測結(jié)果顯示:ITGA5蛋白質(zhì)在肝癌組織中的相對表達量(0.583±0.041)高于癌旁組織中的相對表達量(0.048±0.029);caspase 3蛋白質(zhì)在肝癌組織中的相對表達量(0.168±0.072)低于癌旁組織中的表達量(3.885±0.629)。差異均有統(tǒng)計學意義(P均0.01)。4.肝癌組織中,ITGA5與caspase 3的表達呈負相關(guān)(r=-0.341,P0.05)。結(jié)論ITGA5在肝癌組織中表達增高,且和caspase 3的表達呈負相關(guān),并與肝癌的TNM分期及腫瘤轉(zhuǎn)移有關(guān),提示其表達的高低可能與肝癌轉(zhuǎn)移有關(guān)。
