【摘要】：目的：檢測(cè)c-Myc和VEGF-A在胃癌中的表達(dá)水平及其相關(guān)性,并分析其與胃癌臨床病理參數(shù)之間的關(guān)系。方法：1、應(yīng)用免疫組織化學(xué)方法檢測(cè)90例胃癌組織及其癌旁正常胃組織中c-Myc和VEGF-A蛋白的表達(dá)。2、采用qRT-PCR和Western blotting法對(duì)20例新鮮收集的胃癌組織及其癌旁正常胃組織中c-Myc和VEGF-AmRNA與蛋白的表達(dá)進(jìn)行檢測(cè)。結(jié)果：1、免疫組織化學(xué)結(jié)果顯示：c-Myc在胃癌組織中表達(dá)陽(yáng)性率為61.1%(55/90),在癌旁正常胃組織中為27.8%(25/90),二者之間的差異具有統(tǒng)計(jì)學(xué)意義(P0.05)；VEGF-A在胃癌組織中表達(dá)陽(yáng)性率為68.9%(62/90),在癌旁正常胃組織中為26.7%(24/90),二者之間的差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。2、c-Myc蛋白的表達(dá)與浸潤(rùn)深度、腫瘤分期及淋巴結(jié)轉(zhuǎn)移明顯相關(guān)(P0.05),而與年齡、性別、腫瘤的大小和部位及分化程度無(wú)關(guān)(P0.05);VEGF-A蛋白的表達(dá)與浸潤(rùn)深度、分化程度、腫瘤分期及淋巴結(jié)轉(zhuǎn)移明顯相關(guān)(P0.05),而與年齡、性別、腫瘤的大小和部位無(wú)關(guān)(P0.05)。3、相關(guān)分析結(jié)果顯示,c-Myc和VEGF-A的表達(dá)呈正相關(guān)(P0.05)。4、qRT-PCR結(jié)果顯示：c-Myc和VEGF-A mRNA在胃癌組織中的表達(dá)明顯高于癌旁正常胃組織,差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。5、Western-Blotting的結(jié)果顯示：c-Myc和VEGF-A在胃癌組織中的的表達(dá)明顯高于癌旁正常胃組織,差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論：1、c-Myc和VEGF-A的表達(dá)和與胃癌的浸潤(rùn)深度、淋巴結(jié)轉(zhuǎn)移和臨床分期相關(guān)。2、c-Myc和VEGF-A在胃癌組織中的表達(dá)明顯高于癌旁正常胃組織,且兩者具有協(xié)同作用,在胃癌的發(fā)生、發(fā)展過(guò)程中起重要作用。
[Abstract]:Objective: to detect the expression of c-Myc and VEGF-A in gastric cancer and their correlation, and to analyze the relationship between c-Myc and VEGF-A and the clinicopathological parameters of gastric cancer. The expression of c-Myc and VEGF-A protein in 90 cases of gastric cancer and its adjacent normal gastric tissues were detected by immunohistochemical method. Adopting qRT-PCR and Western. The expression of c-Myc, VEGF-AmRNA and protein in 20 freshly collected gastric cancer tissues and their adjacent normal gastric tissues were detected by blotting. The results of immunohistochemistry showed that the positive rate of the expression of w c-Myc in gastric cancer was 61.1% 55 / 90, and in the adjacent normal gastric tissue was 27.8 / 90). The difference between them was statistically significant (P 0.05). The positive rate of VEGF-A expression in gastric cancer tissues was 68.9 / 90, and that in adjacent normal gastric tissues was 26.724 / 90). The difference between the two groups was statistically significant (P 0.05). The expression of c-Myc protein was significantly correlated with the depth of invasion, tumor stage and lymph node metastasis, but with age and sex. The size, location and differentiation of the tumor were not related to P0.05; The expression of VEGF-A protein was significantly correlated with depth of invasion, degree of differentiation, tumor stage and lymph node metastasis (P 0.05), but with age and sex. The expression of c-Myc and VEGF-A was positively correlated with that of P0.054.The size and location of the tumor were not related to the size and location of the tumor. The correlation analysis showed that there was a positive correlation between the expression of c-Myc and VEGF-A. The results of qRT-PCR showed that the expression of c-Myc and VEGF-A mRNA in gastric cancer was significantly higher than that in normal gastric tissues adjacent to cancer, and the difference was statistically significant (P 0.05). The results of Western-Blotting showed that the expression of c-Myc and VEGF-A in gastric cancer was significantly higher than that in adjacent normal gastric tissues. Conclusion the expression of c-Myc and VEGF-A and the expression of c-Myc and VEGF-A are related to the depth of invasion, lymph node metastasis and clinical stage of gastric cancer. The expression of c-Myc and VEGF-A in gastric cancer tissues was significantly higher than that in adjacent normal gastric tissues, and both had synergistic effects, which played an important role in the occurrence and development of gastric cancer.
