電針對淀粉樣前體蛋白轉(zhuǎn)基因小鼠海馬微血管淀粉樣沉積的影響及其與低密度脂蛋白相關(guān)受體1的關(guān)系

【摘要】:目的:探討電針治療阿爾茨海默病的可能作用機制。方法:將13月齡的淀粉樣前體蛋白(APP)轉(zhuǎn)基因小鼠隨機分為模型組和電針治療組,以相同月齡和背景的C 57 BL/6小鼠作對照組。電針治療組取"百會""涌泉"穴,每次留針15 min,隔日1次,針刺3個月。以LashleyⅢ水迷宮測定各組小鼠的學習記憶能力,以免疫組化方法檢測海馬β淀粉樣蛋白(amyloidβprotein,Aβ)、低密度脂蛋白相關(guān)受體1(low densi-ty lipoprotein receptor-related protein 1,LRP 1)的表達。結(jié)果:模型組水迷宮游出時間較對照組延長(P0.05),海馬溝微血管壁Aβ1-42表達的累積吸光度較對照組升高(P0.01),海馬溝微血管壁LRP 1表達較對照組減低(P0.01);而電針治療組的水迷宮游出時間明顯低于模型組,海馬溝微血管壁Aβ1-42表達低于模型組,海馬溝微血管壁LRP 1表達高于模型組(均P0.05)。結(jié)論:電針治療可能通過提高腦微血管壁Aβ受體LRP 1的清除轉(zhuǎn)運能力,降低APP轉(zhuǎn)基因鼠腦微血管壁的Aβ沉積,從而改善其學習、記憶能力。
[Abstract]:Objective: to explore the possible mechanism of electroacupuncture in the treatment of Alzheimer's disease. Methods: the 13 month old transgenic mice with amyloid precursor protein (APP) were randomly divided into two groups: model group and electroacupuncture group. C57 BL/6 mice of the same age and background were used as the control group. In the electroacupuncture treatment group, the points of "Baihui" and "Yongquan" were taken for 15 mins each time, once every other day, and needled for 3 months.
