【摘要】： 第一部分SLE小鼠模型的建立 目的:探討腹腔注射細(xì)菌脂多糖(LPS)制造SLE小鼠模型的最佳劑量。方法:80只小鼠隨機(jī)分為4組,分別腹腔注射LPS 5 mg·kg - 1、2.5 mg·kg - 1和1.25 mg·kg - 1和生理鹽水。結(jié)果:腹腔注射LPS 5 mg·kg - 1、2.5 mg·kg - 1組動(dòng)物腎臟病理改變符合SLE病理特點(diǎn),動(dòng)物24小時(shí)尿蛋白含量、血清ANA的滴度、腎臟免疫復(fù)合物光密度均明顯升高,而腹腔注射LPS 5 mg·kg - 1組動(dòng)物死亡率明顯升高。結(jié)果:腹腔注射LPS 5 mg·kg- 1、2.5 mg·kg - 1均能制造出良好的狼瘡性腎炎模型,而腹腔注射LPS 5 mg·kg- 1動(dòng)物死亡率高。 第二步分中西醫(yī)結(jié)合治療對(duì)SLE模型鼠療效觀察 目的:探討中西醫(yī)結(jié)合治療對(duì)SLE模型鼠的療效。方法:120只小鼠腹腔注射2.5 mg·kg - 1制造SLE模型鼠,隨機(jī)分為6組,分別給予蒸餾水、中藥清熱解毒補(bǔ)腎方、強(qiáng)的松、環(huán)磷酰胺、清熱解毒補(bǔ)腎方+強(qiáng)的松、清熱解毒補(bǔ)腎方+環(huán)磷酰胺在造模的同時(shí)灌胃治療15天。結(jié)果:與對(duì)照組相比,各治療組動(dòng)物死亡率、24小時(shí)尿蛋白含量、血清ANA的滴度、腎小球免疫復(fù)合物沉積均有不同程度的下降,以中西醫(yī)結(jié)合治療組降低最為明顯。結(jié)論:中藥清熱解毒補(bǔ)腎方及西藥強(qiáng)的松、環(huán)磷酰胺對(duì)狼瘡樣小鼠模型均有良好的治療作用,而中西醫(yī)結(jié)合治療效果最好。
[Abstract]:Part I Establishment of SLE Mouse Model. Objective: to study the optimal dosage of intraperitoneal injection of lipopolysaccharide (LPS) to make SLE mouse model. Methods: 80 mice were randomly divided into 4 groups. LPS 5 mg 路kg -1 and 1.25 mg 路kg -1 and normal saline were injected intraperitoneally respectively. Results: the pathological changes of kidney in LPS 5 mg 路kg -1 + 2.5 mg 路kg -1 group were in accordance with the pathological characteristics of SLE. The urine protein content and the titer of serum ANA were observed in 24 hours after intraperitoneal injection of LPS 5 mg 路kg -1 and 2.5 mg 路kg -1 LPS, respectively. The light density of renal immune complex was significantly increased, but the mortality of LPS 5 mg 路kg -1 group was significantly increased. Results: intraperitoneal injection of LPS 5 mg 路kg -1 and 2.5 mg 路kg -1 could produce a good model of lupus nephritis. However, intraperitoneal injection of LPS 5 mg 路kg -1 resulted in high mortality. The second step is to observe the therapeutic effect of integrated traditional Chinese and western medicine on SLE model rats. Objective: to investigate the therapeutic effect of integrated traditional Chinese and western medicine on SLE model mice. Methods: SLE model mice were induced by intraperitoneal injection of 2.5 mg 路kg -1 in 1: 120 mice. The mice were randomly divided into six groups: distilled water, traditional Chinese medicine to clear heat and detoxify kidney, prednisone, cyclophosphamide. Results: compared with the control group, the 24 hour urine protein content and serum ANA titer of each treatment group were compared with those of the control group, which were treated by oral administration of cyclophosphamide at the same time. The deposition of glomerular immune complex decreased in varying degrees, especially in the treatment group of integrated traditional Chinese and western medicine. Conclusion: the traditional Chinese medicine for clearing heat and detoxifying kidney and the western medicine prednisone and cyclophosphamide have good therapeutic effects on the model of lupus like mice. And the combination of traditional Chinese and western medicine treatment effect is the best.
