【摘要】：目的探究腦皮質(zhì)內(nèi)皮屏障抗原(EBA)及血管內(nèi)皮生長因子(VEGF)在放射性血腦屏障損害條件下的動態(tài)變化規(guī)律,為臨床提供參考。方法使用隨機(jī)數(shù)字表法將48只清潔級雄性SD大鼠分為對照組和放射性腦損傷后7、14和28 d 4組,每組12只。采用X線電子計算機(jī)斷層掃描設(shè)備制備大鼠放射性腦損傷模型。大鼠按照3 m L/kg的劑量尾靜脈注射3%伊文思藍(lán)(EB),開顱并暴露腦皮質(zhì)血管,放于微循環(huán)顯微鏡下觀察EB滲出情況,并借助微循環(huán)顯微成像系統(tǒng)評估血腦屏障通透性;使用免疫組化染色的方法來檢測各組大鼠腦皮質(zhì)EBA和VEGF的表達(dá)。結(jié)果與對照組相比,損傷組大鼠腦皮質(zhì)微血管EB外滲量和VEGF的表達(dá)水平于傷后7、14、28 d均有不同程度升高(均P0.05),從傷后7 d至28 d逐漸降低,損傷各亞組之間差異均有統(tǒng)計學(xué)意義(均P0.05),二者呈正相關(guān)(r=0.898,P0.001);EBA表達(dá)水平于傷后各個時間點(diǎn)均下降(均P0.05),從傷后7 d至28 d逐漸升高,損傷各亞組之間差異均有統(tǒng)計學(xué)意義(均P0.05),與EB外滲量呈負(fù)相關(guān)(r=-0.866,P0.001)。結(jié)論大鼠放射性腦損傷后血腦屏障通透性增加與EBA表達(dá)減少、VEGF表達(dá)增加具有重要關(guān)聯(lián)性。
[Abstract]:Objective to explore the dynamic changes of cerebral cortical endothelial barrier antigen (EBA) and vascular endothelial growth factor (VEGF) under the damage of the radiation blood brain barrier (BBB), and to provide reference for clinical practice. Methods 48 male SD rats were divided into control group and 4 groups of 7,14 and 28 d after radiation brain injury by random digital table method, and 12 rats in each group were treated with X ray. The rat model of radiation brain injury was prepared by a computer tomography device. Rats were injected with 3% Evans blue (EB) in the tail vein of 3 m L/kg, craniotomy and exposure of cerebral cortex vessels. The permeability of EB was observed under microcirculation microscope, and the permeability of blood brain barrier was evaluated with microcirculation microimaging system; immunohistochemical staining was used. The expression of EBA and VEGF in cerebral cortex of rats in each group was detected by color method. Results compared with the control group, the EB exudation and the expression level of VEGF in the cerebral cortex microvessels of the injured rats were elevated in different degrees (P0.05), gradually decreased from 7 d to 28 d after injury, and the differences were statistically significant (all P0.05), There was a positive correlation between the two (r=0.898, P0.001), and the expression level of EBA decreased at all time points after injury (all P0.05), and gradually increased from 7 d to 28 d after injury. The differences were statistically significant (P0.05), and negative correlation with EB exudation (r=-0.866, P0.001). Conclusion the permeability of blood brain barrier and EBA table after radiation brain injury in rats The increased expression of VEGF has an important correlation.
