成骨細(xì)胞MC3T3-E1對高重力的力學(xué)生物學(xué)響應(yīng)

【摘要】:目的探討高重力對前成骨細(xì)胞MC3T3-E1形態(tài)和成骨功能的影響。方法將MC3T3-E1細(xì)胞按不同高重力分為對照組、5 g組、10 g組、15 g組、20 g組,實(shí)驗(yàn)組每次加載30 min連續(xù)加載3 d,對照組同步置于除g值外相同的環(huán)境中。鬼筆環(huán)肽染色考察細(xì)胞骨架蛋白形態(tài);堿性磷酸酶(alkaline phosphatase,ALP)活性檢測試劑盒檢測ALP含量;實(shí)時(shí)定量PCR檢測ALP、Ⅰ型膠原(collagenⅠ,ColⅠ)、骨鈣素(osteocalcin,OC)、runt相關(guān)轉(zhuǎn)錄因子(runt-related transcription factors,Runx2)的基因表達(dá);Western blotting檢測ColⅠ、OC蛋白表達(dá)。結(jié)果高重力條件下成骨細(xì)胞胞體變薄,細(xì)胞表面積明顯增大;細(xì)胞骨架排列結(jié)構(gòu)變得疏松、松散,微絲肌動蛋白束狀結(jié)構(gòu)減少,微絲呈彌散狀,有序性降低。成骨分化相關(guān)指標(biāo)ALP、ColⅠ、OC、Runx2的基因均顯著上調(diào);ColⅠ和OC蛋白亦呈現(xiàn)高表達(dá)。對照組茜素紅染色只有極少量面積很小的桔紅色結(jié)節(jié),高重力加載后各組細(xì)胞明顯形成面積大小不一的桔紅色結(jié)節(jié)。結(jié)論高重力下成骨細(xì)胞可通過骨架結(jié)構(gòu)發(fā)生重排引起細(xì)胞形態(tài)的改變,并進(jìn)一步通過上調(diào)分化相關(guān)基因和蛋白表達(dá),有效刺激成骨細(xì)胞成熟分化。
[Abstract]:Objective to investigate the effects of high gravity on the morphology and osteogenic function of proosteoblasts (MC3T3-E1).Methods MC3T3-E1 cells were divided into control group (5 g), 10 g group (15 g) and 20 g group according to different high gravity.
