3-溴丙酮酸對(duì)秀麗隱桿線蟲(chóng)的毒性和代謝的影響(英文)

目的:通過(guò)3-溴丙酮酸(3-BrPA)處理秀麗隱桿線蟲(chóng)(Caenorhabditis elegans),觀察3-BrPA對(duì)線蟲(chóng)的毒性和生存周期的影響。通過(guò)秀麗隱桿線蟲(chóng)RNA干擾(RNAi)和突變株,分析3-BrPA對(duì)線蟲(chóng)糖酵解途徑己糖激酶家族和藥物代謝的關(guān)鍵酶細(xì)胞色素P450 (cytochrome P450,CYP)家族的影響。創(chuàng)新點(diǎn):首次報(bào)道了3-BrPA對(duì)秀麗隱桿線蟲(chóng)有毒性作用,己糖激酶是3-BrPA對(duì)線蟲(chóng)作用的重要靶點(diǎn),而CYP-35A家族是線蟲(chóng)代謝3-BrPA的主要酶類。方法:用不同濃度的3-BrPA處理秀麗隱桿線蟲(chóng),每24 h監(jiān)測(cè)一次存活率;用實(shí)時(shí)熒光定量聚合酶鏈反應(yīng)(qPCR)檢測(cè)代謝相關(guān)基因的表達(dá);通過(guò)RNAi沉默己糖激酶家族基因hxk-7、hxk-2和hxk-3;計(jì)算3-BrPA處理hxk家族RNAi株和細(xì)胞色素P450cym-35家族突變株后的致死中濃度(LC50)。結(jié)論:3-BrPA對(duì)線蟲(chóng)有明顯的毒性效應(yīng)(圖1);與對(duì)照組比較,3-BrPA處理組的線蟲(chóng)平均壽命明顯縮短(圖2);3-BrPA處理線蟲(chóng)后hxk-1、hxk-2和hxk-3的信使RN...
