馬尾松花粉提取物對前列腺肥大小鼠的影響

【摘要】:前列腺肥大(BPH)是老年男性的常見疾病,為患者的身體和心靈造成了很大的損害。目前治療BPH的主流藥物為5α-還原酶抑制劑和α1-腎上腺素受體阻滯劑,但它們均存在一定的副作用。馬尾松花粉提取物(MPPE)是一種具有良好的抗炎和抗氧化活性的天然植物提取物,其營養(yǎng)豐富,無毒副作用。很多研究表明,MPPE可用于改善BPH。本實驗擬采用腹腔注射丙酸睪酮(5mg/kg·d)的方法建立小鼠BPH模型,通過探究MPPE以及SQ、PGY兩種復(fù)方對小鼠BPH的影響,為松花粉進一步開發(fā)為抗BPH藥物或保健品提供理論依據(jù)。實驗包括以下幾部分: 第一,,MPPE對小鼠BPH的預(yù)防作用探究:實驗周期為6周,前兩周三個松花粉組小鼠預(yù)先灌胃相應(yīng)劑量的MPPE。后四周,在腹腔注射丙酸睪酮建立BPH模型的同時給予三種不同劑量(20、40、80mg/kg)的MPPE。實驗結(jié)束時收集小鼠尿液,進行代謝組學(xué)分析,然后處死小鼠收集血清測定相關(guān)生理指標(biāo),剖取前列腺制作石蠟切片,觀察組織形態(tài)變化。 結(jié)果顯示,與空白組相比,模型組小鼠的前列腺重量、前列腺指數(shù)顯著增加,前列腺上皮細胞增生,向腺腔內(nèi)折疊呈絨毛狀,形態(tài)結(jié)構(gòu)具有明顯的BPH特征。三種劑量的MPPE作用效果有一定差異。與模型組相比,20mg/kg MPPE保護作用較小,而40、80mg/kgMPPE組的前列腺指數(shù)、炎性細胞因子含量、DHT濃度及PACP、NOS的活性均有明顯的降低,NO的生成也相應(yīng)減少。其中,80mg/kg的MPPE效果最好。PLS-DA結(jié)果與生化指標(biāo)基本一致,對照組與模型組小鼠完全分開,說明BPH小鼠的尿液代謝物發(fā)生了明顯的變化。非那雄胺組最接近空白組,MPPE的保護作用比較明顯,但并未使小鼠完全恢復(fù)健康。 第二,MPPE對小鼠BPH的治療作用探究:與預(yù)防作用部分實驗不同的是,治療作用實驗先腹腔注射丙酸睪酮4周建立BPH模型。然后再分別灌胃不同濃度的MPPE治療4周。結(jié)果顯示,模型組小鼠的前列腺重量、前列腺指數(shù)顯著增加,前列腺組織形態(tài)具有明顯的BPH特征。20mg/kg MPPE作用較小,80mg/kg的MPPE效果最好。BPH小鼠的尿液代謝物發(fā)生了明顯的變化,MPPE作用效果比較明顯,但并未使小鼠完全恢復(fù)健康。非那雄胺對BPH的改善效果最佳。 另外,對PI百分抑制率進行對比,預(yù)防試驗中三個劑量MPPE的抑制率分別為26.09%、52.17%、69.57%,而治療實驗的則分別為33.33%、57.14%、76.19%,可以看出MPPE對BPH的預(yù)防作用優(yōu)于其治療作用。 第三,MPPE及SQ、PGY復(fù)方對小鼠BPH的影響:實驗具體步驟與MPPE治療作用實驗一致,1-4周:建模;5-8周:治療。實驗結(jié)束后測定相關(guān)指標(biāo)。實驗結(jié)果顯示,SQ、PGY兩個復(fù)方的抗BPH活性明顯優(yōu)于MPPE。且SQ的效果比PGY稍好,但兩者差異沒有統(tǒng)計學(xué)意義。 以上結(jié)果表明,(1)MPPE對BPH的預(yù)防作用優(yōu)于其治療作用,推測可能的原因是因為一邊建模一邊給藥,MPPE及時的發(fā)揮了保護作用,減少了小鼠患BPH的可能;(2)SQ、PGY復(fù)方的效果優(yōu)于MPPE,且SQ的效果比PGY稍好。說明SQ、PGY兩種中草藥與MPPE復(fù)合后可以起到一定的強化作用,能更好的改善BPH病癥,對機體產(chǎn)生顯著的保護作用;(3)MPPE可以降低BPH病理癥狀及相關(guān)生化指標(biāo),調(diào)節(jié)標(biāo)志性代謝物的水平,且具有一定的劑量依賴性,MPPE的抗BPH作用機理可能與其抗炎抗氧化活性有關(guān)。
