壬基酚和辛基酚聯(lián)合暴露的血清代謝特征及生物標(biāo)志物

【摘要】:運(yùn)用代謝組學(xué)方法分析了壬基酚(NP)和辛基酚(OP)聯(lián)合暴露后大鼠血清代謝成分變化,雄性SD大鼠隨機(jī)分為對照組、壬基酚和辛基酚(聯(lián)合染毒低劑量組(25mg/kg NP+25mg/kg OP)及高劑量組(75mg/kg NP+75mg/kg OP).連續(xù)灌胃7d后處死.通過超高效液相色譜-飛行時間質(zhì)譜(UPLC/QTOF-MS)技術(shù)獲得大鼠血清的代謝指紋圖譜,通過主成分分析觀察了聯(lián)合染毒的劑量-效應(yīng),通過t檢驗(yàn)篩選潛在的生物標(biāo)志物,并結(jié)合代謝物數(shù)據(jù)庫檢索對潛在的生物標(biāo)志物進(jìn)行鑒定.結(jié)果顯示,對照組、高、低劑量組各組間代謝譜有明顯差別,且隨著染毒劑量的增加毒性增強(qiáng),表現(xiàn)為劑量-效應(yīng)關(guān)系.聯(lián)合染毒后的血清代謝物中,含量升高的有牛黃膽酸及1-棕櫚酰基磷脂酰膽堿,含量降低的有酪氨酸、5-尿苷一磷酸及硫腦苷酯,提示染毒可能對內(nèi)分泌系統(tǒng)、心血管系統(tǒng)、神經(jīng)系統(tǒng)以及核苷酸合成、糖代謝、磷脂代謝等產(chǎn)生不良影響.
[Abstract]:The changes of serum metabolic components of nonylphenol (NP) and octylphenol (OP) after combined exposure were analyzed by metabolomics. Male SD rats were randomly divided into control group, nonylphenol and octylphenol (25 mg/ kg NP + 25 mg/ kg OP) and high-dose group (75 mg/ kg NP + 75 mg/ kg OP). The rats were sacrificed for 7 days. The metabolic fingerprint of rat serum was obtained by ultra-high performance liquid chromatography-time-of-flight mass spectrometry (UPLC/ QTOF-MS), and the dose-effect of the combined exposure was observed by the principal component analysis. The potential biomarkers were screened by t-test. And the potential biomarkers are identified in conjunction with the metabolite database search. The results showed that the metabolic profiles of the control group, the high and low dose group were significantly different, and the dose-effect relationship was observed with the increase of the toxicity of the exposure dose. In the serum metabolites after the combined exposure, the content of the increase in the content of the bezoar cholic acid and the 1-palmatine-based phospholipid and the choline, the content of which is reduced, is tyrosine,5-urea-1-phosphate and a thionyl ester, suggesting that the exposure can be carried out on the endocrine system, the cardiovascular system, the nervous system and the nuclear-acid-acid synthesis, Sugar metabolism, phospholipid metabolism, etc. have adverse effects.
