壬基酚對(duì)雄性小鼠生精能力及睪酮的影響

【摘要】:[目的]研究壬基酚對(duì)雄性小鼠生精能力及血清性激素睪酮的影響。[方法]雄性昆明種小鼠40只,隨機(jī)分為4組,每組10只,分別為壬基酚25、50、100 mg/kg劑量染毒組和溶劑對(duì)照組(玉米油)。各組隔日灌胃給予相應(yīng)劑量的壬基酚溶液,連續(xù)35 d后處死小鼠取血,用酶聯(lián)免疫法測(cè)定小鼠血清睪酮的水平。取左側(cè)睪丸做組織病理切片,取雙側(cè)附睪制備精子懸液,并在光學(xué)顯微鏡下進(jìn)行精子計(jì)數(shù),計(jì)算活精子率和精子畸形率。[結(jié)果]35 d后,與對(duì)照組相比,100 mg/kg劑量染毒組小鼠睪丸組織病理學(xué)切片顯示曲細(xì)精管萎縮,體重減輕幅度升高,睪丸重量下降,精子數(shù)量、精子活動(dòng)度均降低,精子畸形率增高(均P0.05)。染毒組小鼠血清睪酮含量、精子數(shù)量與壬基酚濃度存在明顯劑量-反應(yīng)關(guān)系(r=-0.98,r=-0.96;P0.05)。[結(jié)論]壬基酚對(duì)雄性小鼠生殖系統(tǒng)有一定損害作用,高劑量的壬基酚能損傷小鼠的生精功能。
[Abstract]:Objective to study the effect of nonylphenol on spermatogenesis and serum sex hormone testosterone in male mice. [methods] 40 male Kunming mice were randomly divided into 4 groups with 10 mice in each group: nonylphenol 25, 50100 mg/kg dose group and solvent control group (corn oil). The mice in each group were given the corresponding dose of nonylphenol solution intragastrically every other day. After 35 days, the mice were killed and the serum testosterone level was measured by enzyme-linked immunosorbent assay (Elisa). The left testes were taken as pathological sections, the bilateral epididymis was taken to prepare sperm suspension, and the sperm count was carried out under optical microscope to calculate the live sperm rate and sperm deformity rate. [results] after 35 days, compared with the control group, the pathological sections of testicular histology in the 100 mg/kg dose group showed that the seminal tubules were atrophied, the weight loss was increased, the testicular weight was decreased, the sperm count and sperm motility were decreased, and the sperm deformity rate was increased (P 0.05).
