余氯對(duì)河流水體微生物滅活效應(yīng)的評(píng)價(jià)

【摘要】:【目的】研究不同余氯濃度和暴露時(shí)間對(duì)細(xì)菌的去除效果,分析不同余氯條件對(duì)細(xì)胞ATP的影響!痉椒ā恳院铀形⑸锶郝錇樵囼(yàn)對(duì)象,利用流式細(xì)胞術(shù)(Flow cytometry,FCM)評(píng)估不同余氯濃度和暴露時(shí)間的滅活效果,檢測(cè)不同余氯濃度時(shí)細(xì)胞內(nèi)(外)ATP的變化情況!窘Y(jié)果】不同余氯濃度和暴露時(shí)間對(duì)細(xì)菌的去除效果產(chǎn)生不同的結(jié)果。在余氯濃度2 mg/L情況下,延長(zhǎng)氯暴露時(shí)間可以增加細(xì)菌的去除效果,在余氯濃度≥2 mg/L條件下,較短氯暴露時(shí)間就可以滅活90%細(xì)菌。高核苷酸細(xì)菌(HNA)和低核苷酸細(xì)菌(LNA)表現(xiàn)出不同氯耐受能力,且HNA細(xì)菌相比LNA細(xì)菌較容易受到氯的損傷。細(xì)胞內(nèi)ATP隨余氯濃度增加而減少,在高濃度余氯條件下(≥2 mg/L)細(xì)胞外ATP才會(huì)增加!窘Y(jié)論】微生物活性隨著余氯作用的增加而降低,FCM法和ATP檢測(cè)法可以用于評(píng)估加氯消毒對(duì)微生物穩(wěn)定性的影響。
[Abstract]:[Objective] To study the effect of different residual chlorine concentration and exposure time on the bacterial removal, and to analyze the effect of different residual chlorine on the cell ATP. [Methods] The effect of different residual chlorine concentration and time of exposure was assessed by flow cytometry (FCM), and the change of intracellular (external) ATP was detected by flow cytometry (FCM). [Results] The effects of different residual chlorine concentration and exposure time on the bacteria removal were different. Under the condition of the residual chlorine concentration of 2 mg/ L, the removal effect of bacteria could be increased. Under the condition of the residual chlorine concentration of 2 mg/ L,90% of the bacteria could be inactivated by the shorter chlorine exposure time. The high-nucleonic acid bacteria (HNA) and the low-nuclear-acid bacteria (LNA) exhibited different chlorine tolerance, and the HNA bacteria were more susceptible to chlorine damage than the LNA bacteria. The intracellular ATP decreased with the increase of the residual chlorine concentration, and the ATP of the cells (2 mg/ L) increased with the increase of the residual chlorine concentration. [Conclusion] The microbial activity decreases with the increase of the residual chlorine, and the FCM method and the ATP detection method can be used to evaluate the effect of chlorination disinfection on the microbial stability.
