偏二甲肼對雄性小鼠生殖損傷的研究

【摘要】:目的 研究偏二甲肼對雄性小鼠是否具有生殖損傷,探討偏二甲肼作用下小鼠精子質(zhì)量和活力以及遺傳改變并分析其可能的原因。 方法 實(shí)驗(yàn)選擇健康成熟雄性昆明小鼠20只,隨機(jī)分為4組,每組5只,即:正常對照組(NS組),偏二甲肼(UDMH)低劑量染毒組20mg/kg、中劑量染毒組40mg/kg、高劑量染毒組80mg/kg,稱重,灌胃染毒,2次/周,連續(xù)8周。染毒結(jié)束次日,頸椎脫臼處死小鼠,分別制備檢測樣本,進(jìn)行下列實(shí)驗(yàn):精子質(zhì)量檢測以及精子活力檢測;單細(xì)胞凝膠電泳實(shí)驗(yàn)。 結(jié)果 精子活力檢測結(jié)果顯示,和對照組相比,偏二甲肼作用下精子活力與對照組相比改變不大,未見精子明顯活力減弱現(xiàn)象(P0.05);單細(xì)胞凝膠電泳實(shí)驗(yàn)中,精子細(xì)胞被染成橘紅色,,大部分呈正常形態(tài)且細(xì)胞核無拖尾現(xiàn)象,不同劑量組之間,拖尾細(xì)胞數(shù)差異不明顯(P0.05)。 結(jié)論 未見偏二甲肼對小鼠精子數(shù)量減少和活力減低有明顯相關(guān)。單細(xì)胞凝膠電泳(SCGE)實(shí)驗(yàn)未見偏二甲基肼對小鼠精子細(xì)胞DNA損傷有明顯相關(guān)。
[Abstract]:Aim to study the reproductive damage of unsymmetrical dimethylhydrazine (UDMH) in male mice, to explore the sperm quality, motility and genetic changes under the action of UDMH and to analyze its possible causes. Methods Twenty healthy adult male Kunming mice were randomly divided into 4 groups: normal control group (NS group), unsymmetrical dimethylhydrazine (UDMH) low dose group (20mg / kg), middle dose group (40mg / kg), unsymmetrical dimethylhydrazine (UDMH) group (20 mg / kg) and middle dose group (40 mg / kg / kg). The high dose group was given 80 mg / kg, weighing, twice a week for 8 weeks. The mice were killed the next day after exposure to dislocated cervical vertebra. The test samples were prepared and the following experiments were carried out: sperm quality test and sperm motility test; single cell gel electrophoresis (SCGE) test. Results the results of sperm motility test showed that compared with the control group, the sperm motility under the action of unsymmetrical dimethylhydrazine had no significant change compared with the control group, and there was no obvious decrease in sperm motility (P0.05). In single cell gel electrophoresis (SCGE), sperm cells were stained orange-red, most of them were in normal shape and the nucleus had no tail-dragging phenomenon. There was no significant difference in the number of tail-tailed cells between different dose groups (P0.05). Conclusion there is no significant correlation between unsymmetrical dimethylhydrazine (UDMH) and decreased sperm motility in mice. Single cell gel electrophoresis (SCGE) showed no significant correlation between unsymmetrical dimethylhydrazine (UDMH) and DNA damage in mouse spermatozoa.
