D-AP5對氟砷聯(lián)合染毒原代培養(yǎng)海馬神經(jīng)元突觸相關(guān)蛋白表達(dá)的影響

【摘要】:[目的]探討谷氨酸NMDA受體拮抗劑D-2-氨基-5-磷酰基戊酸(D-AP5)對氟砷聯(lián)合染毒后原代海馬神經(jīng)元突觸相關(guān)蛋白表達(dá)的影響。[方法]將體外培養(yǎng)的海馬神經(jīng)元分為4組,空白對照組、氟砷聯(lián)合染毒組(0.4μg/m L氟化鈉+0.1μg/m L亞砷酸鈉)、D-AP5組(25μmol/L D-AP5)和D-AP5+氟砷聯(lián)合染毒組(0.4μg/m L氟化鈉+0.1μg/m L亞砷酸鈉+25μmol/L D-AP5)。各組海馬神經(jīng)元置于37℃、含有體積分?jǐn)?shù)5%CO_2培養(yǎng)箱中培養(yǎng)24 h。把培養(yǎng)好的各組海馬神經(jīng)元用CCK8檢測活性,用蛋白免疫印跡法檢測海馬神經(jīng)元中NR2B、SYN及GAP43突觸相關(guān)蛋白的表達(dá)水平。[結(jié)果]與空白對照組[(100.00±0.00)%]比較,氟砷聯(lián)合染毒組[(75.81±6.27)%]、D-AP5+氟砷聯(lián)合染毒組[(81.58±5.20)%]的細(xì)胞存活率均明顯降低(P0.05);與氟砷聯(lián)合染毒組[(75.81±6.27)%]比較,D-AP5組[(98.78±3.53)%]、D-AP5+氟砷聯(lián)合染毒組[(81.58±5.20)%]海馬神經(jīng)元存活率均明顯升高(P0.05)。氟砷聯(lián)合染毒組(0.425±0.06)、D-AP5組(0.469±0.03)、D-AP5+氟砷聯(lián)合染毒組(0.354±0.04)的NR2B相對表達(dá)量較空白對照組(0.572±0.05)均有所下降(P0.05),D-AP5+氟砷聯(lián)合染毒組(0.354±0.04)受體蛋白NR2B表達(dá)較氟砷聯(lián)合染毒組(0.425±0.06)降低(P0.05);氟砷聯(lián)合染毒組與D-AP5+氟砷聯(lián)合染毒組的SYN和GAP43相對表達(dá)量均較空白對照組低(P0.05)。而D-AP5組與D-AP5+氟砷聯(lián)合染毒組的SYN和GAP43表達(dá)較氟砷聯(lián)合染毒組均升高(P0.05)。[結(jié)論]氟砷聯(lián)合染毒能夠下調(diào)突觸相關(guān)蛋白SYN及GAP-43表達(dá)水平,谷氨酸NMDA受體拮抗劑D-AP5可以抑制氟砷聯(lián)合染毒對海馬神經(jīng)元突觸相關(guān)蛋白表達(dá)的影響。
