鋁致大鼠認知能力和全基因組甲基化改變的研究
[Abstract]:Objective to study the effects of aluminum on cognitive ability and whole genome methylation in rats. Methods 72 healthy and clean grade male SD rats were randomly divided into 9 groups according to body mass. The experimental part was as follows: control group (normal saline), low, middle, high aluminum dose group (0.27,0.54 and 1.08 mg/kg); Intervention part: control group (saline), high-dose aluminum group (1.08 mg/kg), high-dose aluminum folic acid low, middle, high-dose group (0.7, 1.5 and 3.4 mg/kg), 8 rats in each group. Rats in each group were intraperitoneally injected with the corresponding dose of maltol aluminum (0. 2 ml/d), and folic acid in the intervention group was given 1 ml/100 / g daily for 60 days. The learning and memory ability of rats was measured by Morris water maze, and the whole genome methylation of rats was determined by ELISA method. Results the results of Morris water maze showed that aluminum exposure could significantly shorten the retention time of the target quadrant, decrease the number of traversing the original platform position, and lower aluminum. There was significant difference between the middle and high dose groups compared with the control group (P0.05). The intervention of folic acid in the high-dose aluminum group could significantly prolong the target quadrant residence time and increase the times of crossing the original platform position. There was a significant difference between the high-dose aluminum group and the high-dose aluminum-folic acid group (P0.05). The results of whole genome methylation assay showed that the methylation rate of the whole genome in 0.54 and 1.08 mg/kg groups was significantly lower than that in the control group (P0.05). The whole genome methylation rate of the high-dose folic acid group was significantly higher than that of the high-dose aluminum group (P 0.05), but no significant difference was found between the high-dose folic acid group and the control group. Conclusion Aluminum can decrease the ability of learning and memory in rats and the methylation rate of whole genome in rats. Folic acid may improve the ability of learning and memory in rats.
