甲基叔丁基醚致人B淋巴母細(xì)胞系DNA損傷和氧化應(yīng)激損傷的研究

【摘要】：[目的]通過(guò)體外實(shí)驗(yàn)研究甲基叔丁基醚(MTBE)染毒對(duì)人B淋巴母細(xì)胞系DNA損傷和氧化應(yīng)激水平的影響。[方法]將處于對(duì)數(shù)生長(zhǎng)期的人B淋巴母細(xì)胞系分別以1.25、2.5、5、10、12.5μmol/L的MTBE溶液染毒,24 h后分別收集細(xì)胞和上清液,用彗星試驗(yàn)檢測(cè)人B淋巴細(xì)胞染毒后的尾部DNA百分比及Olive尾矩;同時(shí),用ELISA試劑盒檢測(cè)細(xì)胞上清液中的丙二醛(MDA)、谷胱甘肽過(guò)氧化物酶(GSH-Px)和8-羥基脫氧鳥苷(8-OHd G)水平,并對(duì)MTBE濃度和這些指標(biāo)之間的關(guān)系進(jìn)行相關(guān)性分析。[結(jié)果]MTBE染毒24 h后,人B淋巴母細(xì)胞系在5、10、12.5μmol/L劑量組的尾部DNA百分比及Olive尾矩均高于對(duì)照組(均P0.01);1.25、2.5、5、12.5μmol/L組8-OHd G水平高于對(duì)照組(P0.05);除10μmol/L組的MDA之外,各組MDA、GSH-Px水平均未見(jiàn)明顯改變。MTBE濃度和尾部DNA百分比及Olive尾矩之間呈正相關(guān)關(guān)系(r=0.685,P=0.000;r=0.615,P=0.000)。[結(jié)論]低劑量MTBE染毒可以誘導(dǎo)人B淋巴母細(xì)胞系發(fā)生DNA損傷,DNA損傷可以作為反映MTBE毒性效應(yīng)的一項(xiàng)較敏感的指標(biāo)。
[Abstract]:[objective] to study the effects of methyl tert-Ding Ji ether (MTBE) on DNA damage and oxidative stress of human B lymphoblast cell line in vitro. [methods] the human B lymphoblastoid cell lines in logarithmic growth phase were exposed to MTBE solution of 1.252.5 渭 mol/L and 1012.5 渭 mol/L respectively. After 24 hours, the cells and supernatants were collected, respectively. The tail DNA percentage and Olive tail moment of human B lymphocytes were detected by comet assay. Meanwhile, the levels of malondialdehyde (MDA), glutathione peroxidase (GSH-Px) and 8-hydroxydeoxyguanosine (8-OHd G) in the supernatant were detected by ELISA kit. The correlation between MTBE concentration and these indexes was analyzed. [results] after 24 hours of MTBE exposure, the percentage of tail DNA and the tail moment of Olive of human B lymphoblastoid cell line were higher than those of the control group (P0.01). The level of 8-OHd G in the 12.5 渭 mol/L group was higher than that in the control group (P0.05). There was no significant change in MDA,GSH-Px level except MDA in 10 渭 mol/L group. There was a positive correlation between MTBE concentration, percentage of DNA in tail and tail moment of Olive. [conclusion] low dose MTBE exposure can induce DNA damage in human B lymphoblastoid cell line, and DNA damage may be a sensitive index to reflect the toxic effect of MTBE.
