三氧化二砷對(duì)小鼠成體神經(jīng)發(fā)生影響的研究

【摘要】:目前成體神經(jīng)干細(xì)胞在臨床應(yīng)用中具有廣闊的前景,對(duì)它的研究一直是國(guó)內(nèi)外神經(jīng)領(lǐng)域研究的熱點(diǎn),大量的形態(tài)學(xué)和行為學(xué)實(shí)驗(yàn)表明成體干細(xì)胞與學(xué)習(xí)記憶密切相關(guān)。近年來(lái)砷中毒對(duì)中樞神經(jīng)系統(tǒng)的損害已引起人們的高度重視,而有關(guān)慢性砷中毒成年小鼠動(dòng)物模型成體神經(jīng)發(fā)生的研究尚未見報(bào)道。本實(shí)驗(yàn)通過建立慢性砷中毒小鼠模型,著重應(yīng)用免疫組化技術(shù)在細(xì)胞水平上研究三氧化二砷中毒對(duì)海馬結(jié)構(gòu)中的齒狀回顆粒下層(sub granular zone, SGZ區(qū))的成體神經(jīng)發(fā)生的影響,包括成體干細(xì)胞的增殖速度和新生細(xì)胞的存活,為闡明成體神經(jīng)發(fā)生與慢性砷中毒及學(xué)習(xí)和記憶的關(guān)系提供必要的理論和實(shí)驗(yàn)基礎(chǔ)。目的:探討三氧化二砷對(duì)小鼠海馬成體神經(jīng)發(fā)生的影響。方法:用C57小鼠建立砷中毒模型,和BrdU(5-溴脫氧尿嘧啶核苷)腹腔注射、活體灌注取腦、常規(guī)冰凍切片等方法,并用BrdU免疫組化染色結(jié)合普通光學(xué)顯微鏡進(jìn)行顯微觀察和照相,計(jì)數(shù)小鼠海馬單位長(zhǎng)度齒狀回內(nèi)顆粒細(xì)胞層中BrdU陽(yáng)性細(xì)胞數(shù)并進(jìn)行統(tǒng)計(jì)學(xué)分析。結(jié)果:BrdU注射后存活24小時(shí)處死的空白對(duì)照組(NS) BrdU陽(yáng)性細(xì)胞平均數(shù)是17.63個(gè),低劑量組(1mg/kg)BrdU陽(yáng)性細(xì)胞平均數(shù)是21.17個(gè),高劑量組(2mg/kg) BrdU陽(yáng)性細(xì)胞平均數(shù)是11.50個(gè);BrdU注射后存活28天處死的空白對(duì)照組(NS) BrdU陽(yáng)性細(xì)胞平均數(shù)是11.40個(gè),低劑量組(1mg/kg) BrdU陽(yáng)性細(xì)胞平均數(shù)是5.40個(gè),高劑量組(2mg/kg) BrdU陽(yáng)性細(xì)胞平均數(shù)是5.57個(gè)。結(jié)論:三氧化二砷在一定的濃度范圍內(nèi),可引起海馬齒狀回成體神經(jīng)干細(xì)胞數(shù)量的增加,促進(jìn)其成體神經(jīng)發(fā)生,但其會(huì)抑制新生細(xì)胞的存活。
[Abstract]:At present, adult neural stem cells have a broad prospect in clinical application. The research on adult neural stem cells has always been a hot spot in the field of nerve research at home and abroad. A large number of morphological and behavioral experiments show that adult neural stem cells are closely related to learning and memory. In recent years, the damage of arsenic poisoning to the central nervous system has attracted great attention, but the study on adult neurogenesis of adult mice model of chronic arsenic poisoning has not been reported. In this study, we established a model of chronic arsenism in mice and studied the effects of arsenic trioxide on the neurogenesis of adult neurons in the subgranular (sub granular zone, SGZ region of dentate gyrus in the hippocampal formation by immunohistochemical technique at the cellular level. The proliferation rate of adult stem cells and the survival of newborn cells provide the necessary theoretical and experimental basis for elucidating the relationship between adult neurogenesis and chronic arsenism, learning and memory. Objective: to investigate the effect of arsenic trioxide on the development of adult hippocampal nerve in mice. Methods: the model of arsenic poisoning in C57 mice was established, and BrdU (5-bromodeoxyuridine) was injected intraperitoneally. The BrdU positive cells in the granular cell layer of the dentate gyrus of the hippocampal unit length of mice were counted and statistically analyzed by BrdU immunohistochemical staining and light microscope. Results: the average number of (NS) BrdU positive cells in the blank control group was 17.63, and that in the low dose group (1mg/kg) was 21.17. The average number of BrdU positive cells in high dose group (2mg/kg) was 11.50; The average number of (NS) BrdU positive cells in the blank control group was 11.40, and that in the low dose group (1mg/kg) was 5.40. The average number of BrdU positive cells in high dose group (2mg/kg) was 5.57. Conclusion: arsenic trioxide can increase the number of adult neural stem cells in dentate gyrus of sea horse in a certain concentration range, and promote the development of adult nerve, but it can inhibit the survival of newborn cells.
