雪蓮果汁對高脂模型大鼠的降脂作用及其機制初步探討

【摘要】:觀察雪蓮果對高脂高糖致高脂大鼠模型的降脂作用,并對其影響機制進行初步探討。取SD雄性大鼠喂養(yǎng)高脂高糖飼料建立高脂模型成功后,將高脂模型大鼠隨機分為陰性對照組,陽性對照組,雪蓮果高、中、低劑量組。各組動物按各藥物和劑量灌胃給藥至第16周后測定血清血脂水平及肝臟AMPK和ACC在基因和蛋白表達水平。與陰性對照相比,高、中劑量組TG、TC及LDL-C含量降低,HDL-C含量升高;AMPK的基因和蛋白表達水平高于陰性對照組;ACC的基因和蛋白表達水平低于陰性組;低劑量組與陰性對照比無明顯差異;一定劑量的雪蓮果汁能上調(diào)大鼠肝臟組織中AMPK m RNA和蛋白質(zhì)表達,下調(diào)ACC m RNA和蛋白質(zhì)表達,從而抑制脂肪酸合成,同時加速脂肪酸氧化,達到降脂作用。
[Abstract]:To observe the hypolipidemic effect of Saussurea nucifera fruit on hyperlipidemic rat model induced by high fat and high sugar, and to explore its mechanism. After SD male rats were fed with high fat and high sugar diet, the rats were randomly divided into negative control group, positive control group, high, middle and low dosage group of Saussurea nucifera fruit. The serum lipids and the expression of AMPK and ACC in the liver were measured by intragastric administration of various drugs and doses until the 16th week. Compared with the negative control group, the content of TG,TC and LDL-C decreased and the content of HDL-C increased in the middle dose group, the gene and protein expression of AMPK was higher than that of the negative control group, the expression level of ACC gene and protein was lower than that of the negative group. There was no significant difference between low dose group and negative control group. A certain dose of Saussurea nucifera juice could up-regulate the expression of AMPK m RNA and protein, down-regulate the expression of ACC m RNA and protein, thus inhibit the synthesis of fatty acids, and accelerate the oxidation of fatty acids to achieve the lipid-lowering effect.
