X射線對(duì)人周圍血淋巴細(xì)胞Gadd45 mRNA表達(dá)影響

【摘要】:目的分析不同受照劑量和不同受照時(shí)間對(duì)人周圍血淋巴細(xì)胞生長(zhǎng)抑制和DNA損傷基因45(Gadd45)mRNA相對(duì)表達(dá)水平的影響,探討Gadd45基因作為生物劑量計(jì)的可能性。方法 1劑量-效應(yīng)關(guān)系研究。以0.00、0.10、0.25、0.50、1.00、2.00、3.00和5.00 Gy劑量X射線離體照射健康人周圍血樣,分離淋巴細(xì)胞進(jìn)行檢測(cè);2時(shí)間-效應(yīng)關(guān)系研究。以2.00 Gy劑量X射線離體照射健康人周圍血樣,分離淋巴細(xì)胞后分別于培養(yǎng)0、3、6、12、24和48 h時(shí)收獲細(xì)胞。采用實(shí)時(shí)熒光定量聚合酶鏈反應(yīng)法檢測(cè)Gadd45 mRNA相對(duì)表達(dá)水平。結(jié)果 1 0.00~0.50 Gy劑量范圍內(nèi),Gadd45 mRNA相對(duì)表達(dá)水平隨照射劑量的增加而增加,回歸方程為y=1.056 9+7.132 2 x[決定系數(shù)(R2)=0.992,P0.01]。0.50 Gy劑量組Gadd45 mRNA相對(duì)表達(dá)水平達(dá)到峰值,為0.00 Gy劑量組的4.53倍;0.50~5.00 Gy劑量范圍內(nèi),Gadd45 mRNA相對(duì)表達(dá)水平呈波動(dòng)性下降。22.00 Gy劑量照射后3~12 h,Gadd45 mRNA相對(duì)表達(dá)水平隨照射時(shí)間的增加而增加,回歸方程為y=-6.366 0+4.965 0 x(R2=0.932,P0.05)。照射后12 h時(shí)間組Gadd45 mRNA相對(duì)表達(dá)水平達(dá)到峰值,為0 h時(shí)間組的57.68倍;Gadd45 mRNA相對(duì)表達(dá)水平在照射后24~48 h大幅度下降,照射后48 h時(shí)間組為0 h時(shí)間組的2.08倍。結(jié)論 X射線照射可致Gadd45 mRNA表達(dá)上調(diào),在一定劑量和時(shí)間范圍內(nèi)具有良好的劑量-效應(yīng)和時(shí)間-效應(yīng)關(guān)系;但Gadd45基因作為新型生物劑量計(jì)仍需進(jìn)一步研究驗(yàn)證。
[Abstract]:Objective to analyze the effects of different doses and time of irradiation on the growth inhibition of human peripheral blood lymphocytes and the relative expression of DNA damage gene 45 (Gadd45) mRNA, and to explore the possibility of Gadd45 gene as a biological dosimeter. Methods 1 the dose-effect relationship was studied. The peripheral blood samples of healthy people were irradiated in vitro with the doses of 0.000.10 ~ 0.25 ~ 0.50 ~ 1.00 ~ 1.00 ~ 2.00 ~ 3.00 and 5.00 Gy respectively, and the relationship between time and effect was studied. Peripheral blood samples of healthy people were irradiated with 2. 00 Gy dose of X ray in vitro. Lymphocytes were isolated and cultured for 24 h and 48 h, respectively. The relative expression of Gadd45 mRNA was detected by real-time fluorescence quantitative polymerase chain reaction. Results within the dose range of 0.50 Gy, the relative expression of Gadd45 mRNA increased with the increase of irradiation dose. The regression equation was 1.056 9 7.132 2 x [determination coefficient (R2) = 0.992 2]. P0.01. The relative expression of Gadd45 mRNA in 0.50 Gy group reached the peak value, 4.53 times of that in 0.00 Gy group. Within the dose range of 0.50 Gy 5.00 Gy, the relative expression level of Gadd45 mRNA decreased, and the relative expression level of Gadd45 mRNA increased with the increase of irradiation time at 3h 12 h after 22.00 Gy dose irradiation. The regression equation was Yi-6.366 0 4.965 x (R _ 2N _ (0.932) P _ (0.05). The relative expression level of Gadd45 mRNA reached the peak at 12 h after irradiation, which was 57.68 times of that in 0 h group, and the relative expression level of Gadd45 mRNA decreased significantly at 24 h after irradiation 48 h after irradiation, and 2.08 times as much as 0 h at 48 h after irradiation. Conclusion the expression of Gadd45 mRNA can be upregulated by X-ray irradiation, which has a good dose-effect and time-effect relationship within a certain dose and time range, but the Gadd45 gene as a new biological dosimeter needs further study and verification.
