水解乳清蛋白對(duì)輻射損傷大鼠蛋白質(zhì)營(yíng)養(yǎng)狀態(tài)及抗氧化能力的影響

【摘要】:目的 乳清蛋白屬于優(yōu)質(zhì)蛋白,是臨床腸內(nèi)營(yíng)養(yǎng)配方食品的優(yōu)質(zhì)蛋白質(zhì)來(lái)源。水解乳清蛋白是乳清蛋白經(jīng)酶促反應(yīng)水解后的產(chǎn)物,所含的小分子短肽和游離氨基酸可在腸道不經(jīng)消化而被直接吸收,較乳清蛋白有更多生物活性。本文通過(guò)就水解乳清蛋白對(duì)輻射損傷大鼠蛋白質(zhì)營(yíng)養(yǎng)狀態(tài)及抗氧化能力影響的研究,旨在為其臨床應(yīng)用提供依據(jù)。 方法 清潔級(jí)SD大鼠40只(雌雄各半),鼠齡14周,體重269.7±15.2g,隨機(jī)分為正常對(duì)照組、實(shí)驗(yàn)對(duì)照組、實(shí)驗(yàn)Ⅰ組、實(shí)驗(yàn)Ⅱ組,每組10只。正常對(duì)照組、實(shí)驗(yàn)對(duì)照組飼料氮源為乳清蛋白,實(shí)驗(yàn)Ⅰ組、實(shí)驗(yàn)Ⅱ組飼料氮源為水解乳清蛋白,實(shí)驗(yàn)Ⅱ組大鼠同時(shí)每天灌胃10%富硒SOD綠茶溶液[茶粉1g/(kg·d)],各組飼料其他組成均參照AIN-76、AIN-93配方配制。實(shí)驗(yàn)對(duì)照組、實(shí)驗(yàn)Ⅰ組和實(shí)驗(yàn)Ⅱ組動(dòng)物給予一次性6Gray劑量的60Co-γ射線全身輻照。記錄每天大鼠的進(jìn)食量,,定期監(jiān)測(cè)體重,21天后收集72小時(shí)糞便,采集血液并處死動(dòng)物,測(cè)定糞氮,計(jì)算蛋白質(zhì)AD值,進(jìn)行血清TP、ALB和MDA含量及T-AOC、SOD、GSH-PX活力等相關(guān)指標(biāo)測(cè)定,并進(jìn)行腸黏膜HE染色。 結(jié)果 1、正常對(duì)照組動(dòng)物整個(gè)實(shí)驗(yàn)期攝食量正常,體重持續(xù)增加。實(shí)驗(yàn)對(duì)照組、實(shí)驗(yàn)Ⅰ組、實(shí)驗(yàn)Ⅱ組動(dòng)物輻照后攝食量和體重均明顯低于正常對(duì)照組(p<0.01),實(shí)驗(yàn)Ⅰ組、實(shí)驗(yàn)Ⅱ組動(dòng)物體重分別在輻照9天后、6天后開始增加,并且實(shí)驗(yàn)Ⅱ組動(dòng)物體重非常顯著高于實(shí)驗(yàn)對(duì)照組(p<0.01),也顯著高于實(shí)驗(yàn)Ⅰ組(p<0.05)。 2、實(shí)驗(yàn)對(duì)照組、實(shí)驗(yàn)Ⅰ組、實(shí)驗(yàn)Ⅱ組糞氮排出量均非常顯著高于正常對(duì)照組(p<0.01),但實(shí)驗(yàn)Ⅰ組、實(shí)驗(yàn)Ⅱ組非常顯著低于實(shí)驗(yàn)對(duì)照組(p<0.01)。在實(shí)驗(yàn)動(dòng)物輻射損傷情況下AD值計(jì)算結(jié)果表明,實(shí)驗(yàn)對(duì)照組、實(shí)驗(yàn)Ⅰ組、實(shí)驗(yàn)Ⅱ非常顯著低于正常對(duì)照組(p<0.01),而實(shí)驗(yàn)Ⅰ組、實(shí)驗(yàn)Ⅱ組非常顯著高于實(shí)驗(yàn)對(duì)照組(p<0.01)。 3、實(shí)驗(yàn)對(duì)照組血清TP和ALB含量均顯著低于正常對(duì)照組(p<0.01)。實(shí)驗(yàn)Ⅰ組、實(shí)驗(yàn)Ⅱ組血清TP、ALB含量雖然低于正常對(duì)照組,但僅血清ALB含量在實(shí)驗(yàn)Ⅰ組與正常對(duì)照組間存在統(tǒng)計(jì)學(xué)差異(p<0.05),并且實(shí)驗(yàn)Ⅰ組、實(shí)驗(yàn)Ⅱ組血清ALB含量非常顯著高于實(shí)驗(yàn)對(duì)照組(p<0.01)。 4、實(shí)驗(yàn)對(duì)照組血清T-AOC、T-SOD、GSH-PX活力均非常顯著低于正常對(duì)照組(p<0.01)。實(shí)驗(yàn)Ⅰ組、實(shí)驗(yàn)Ⅱ組血清T-AOC和GSH-PX活力也非常顯著低于正常對(duì)照組(p<0.01),但非常顯著高于實(shí)驗(yàn)對(duì)照組(p<0.01),并且實(shí)驗(yàn)Ⅱ組血清GSH-PX活力也非常顯著高于實(shí)驗(yàn)Ⅰ組(p<0.01)。實(shí)驗(yàn)對(duì)照組、實(shí)驗(yàn)Ⅰ組、實(shí)驗(yàn)Ⅱ組血清MDA含量均非常顯著高于正常對(duì)照組(p<0.01),但實(shí)驗(yàn)Ⅰ組、實(shí)驗(yàn)Ⅱ組非常顯著低于實(shí)驗(yàn)對(duì)照組(p<0.01)。 5、實(shí)驗(yàn)各組腸黏膜HE染色也顯示不同程度的病理學(xué)改變。 結(jié)論 1、6Gray劑量的60Co-γ射線一次性全身輻照可嚴(yán)重影響受試動(dòng)物營(yíng)養(yǎng)狀態(tài)。 2、水解乳清蛋白的消化、吸收與利用明顯優(yōu)于乳清蛋白,更有利于改善輻射損傷大鼠的蛋白質(zhì)營(yíng)養(yǎng)狀態(tài)。 3、水解乳清蛋白可以減輕輻照大鼠的過(guò)氧化損傷,與富硒SOD綠茶粉聯(lián)合應(yīng)用效果更好。
