慢性砷染毒對(duì)大鼠睪丸肝細(xì)胞生長(zhǎng)因子及其受體c-Met表達(dá)的影響

【摘要】:目的探討砷對(duì)大鼠睪丸中肝細(xì)胞生長(zhǎng)因子(hepatocyte growth factor,HGF)及其受體c-Met表達(dá)的影響。方法將40只健康清潔雄性SD大鼠隨機(jī)分為4組,分別為對(duì)照(蒸餾水)組和2.4、12.0、60.0 mg/L亞砷酸鈉染毒組,每組10只。采用自由飲用方式進(jìn)行染毒,連續(xù)染毒6個(gè)月。測(cè)定大鼠睪丸組織內(nèi)HGF及其受體c-Met的陽(yáng)性表達(dá)及精子的凋亡情況。結(jié)果與對(duì)照組比較,各濃度亞砷酸鈉染毒組大鼠睪丸組織內(nèi)HGF、c-Met的陽(yáng)性表達(dá)水平均較低,差異均有統(tǒng)計(jì)學(xué)意義(P0.05);且隨著亞砷酸鈉染毒濃度的升高,大鼠的HGF、c-Met的陽(yáng)性表達(dá)水平呈下降趨勢(shì)。與對(duì)照組比較,各濃度亞砷酸鈉染毒組大鼠的精子凋亡率均升高,差異有統(tǒng)計(jì)學(xué)意義(P0.05);且隨著亞砷酸鈉染毒濃度的升高,大鼠的精子凋亡率呈上升趨勢(shì)。結(jié)論慢性砷染毒可能通過(guò)抑制HGF-c-Met通路引起大鼠睪丸組織的損傷及精子凋亡。
[Abstract]:Objective to investigate the effect of arsenic on the expression of hepatocyte growth factor (hepatocyte growth factor,HGF) and its receptor c-Met in rat testis. Methods Forty healthy and clean male SD rats were randomly divided into 4 groups: control group (distilled water) and sodium arsenite group (2.40.12.0 mg/L), 10 rats in each group. The free drinking method was used for 6 months. The positive expression of HGF and its receptor c-Met and the apoptosis of spermatozoa were measured in rat testis. Results compared with the control group, the positive expression level of HGF,c-Met in testis of rats exposed to sodium arsenite was lower than that of control group (P0.05). With the increase of sodium arsenite concentration, the positive expression of HGF,c-Met in rats decreased. Compared with the control group, the rate of sperm apoptosis of rats exposed to sodium arsenite was significantly higher than that of the control group (P0.05), and with the increase of the concentration of sodium arsenite, the rate of sperm apoptosis showed an increasing trend. Conclusion chronic arsenic exposure may induce testicular injury and sperm apoptosis in rats by inhibiting HGF-c-Met pathway.
