輻射生物效應(yīng)中微劑量學(xué)細胞模型初步研究

【摘要】:人體輻射健康危害取決于輻射能量在人體的沉積分布及后續(xù)復(fù)雜的生物學(xué)過程。依據(jù)輻射損傷機制經(jīng)典學(xué)說--靶學(xué)說,DNA分子是輻射損傷最重要的靶,輻射能量在其內(nèi)的沉積特點對輻射生物效應(yīng)的解釋具有重要意義?紤]到細胞是人體的基本功能單元且DNA分子受損后的各種生物學(xué)響應(yīng)均與細胞環(huán)境有關(guān),因此,在細胞水平開展輻射劑量學(xué)研究對輻射損傷機制的深入了解及其在輻射防護和放射治療等方面的實用化具有重要意義。 目的:單細胞模型是輻射能量在細胞水平上沉積特點和規(guī)律獲取的重要平臺。本文旨在得到合理可行的單細胞模型建立方法,并據(jù)此建立更加接近真實細胞的Monte Carlo計算模型。 材料與方法:采用具有腫瘤細胞代表性的人口腔上皮癌細胞作為研究對象,基于生物學(xué)手段和計算機圖形技術(shù)摸索模型的建立方法,并結(jié)合GEANT4蒙特卡羅模擬軟件進行輻射照射下細胞內(nèi)能量沉積規(guī)律的獲取。 結(jié)果:成功建立了單細胞模型,并針對該模型進行了適用性和實用性的測試。在此基礎(chǔ)上進行了劑量學(xué)計算,結(jié)果表明,在綜合考慮計算時間和計算精度的情況下對7組數(shù)據(jù)進行分析,最終將圖像分辨率設(shè)為0.3-0.4μm范圍內(nèi),用200個3MeV的α粒子對單細胞模型進行照射,,得到細胞核內(nèi)單個粒子擊中細胞的平均比能均值為0.209Gy;細胞質(zhì)內(nèi)單個粒子擊中細胞的平均比能均值為0.044Gy。 本文得到的計算結(jié)果與已公布數(shù)據(jù)吻合,建立單細胞體素模型的方法切實可行且通用性強;細胞核的密度與細胞形態(tài)對模型計算結(jié)果有重要影響,建立不同細胞系的細胞庫是對于放射生物學(xué)的研究非常有必要的。
[Abstract]:Radiation health hazard depends on the deposition and distribution of radiation energy in human body and subsequent complex biological processes. According to the classical theory of radiation damage mechanism, target theory, DNA molecule is the most important target of radiation damage. Considering that cells are the basic functional units of the human body and that the various biological responses of damaged DNA molecules are related to the cellular environment, Radiation dosimetry at the cellular level is of great significance in understanding the mechanism of radiation damage and in radiation protection and radiation therapy. Objective: single cell model is an important platform for obtaining the characteristics and regularity of radiation energy deposition at cell level. The purpose of this paper is to obtain a reasonable and feasible method for establishing a single cell model, and then to establish a Monte Carlo calculation model which is closer to the real cell. Materials and methods: human oral epithelial cancer cells, which are representative of tumor cells, were used as research objects. GEANT4 Monte Carlo simulation software was used to obtain the intracellular energy deposition under radiation irradiation. Results: a single cell model was successfully established, and the applicability and practicability of the model were tested. On this basis, dosimetric calculations are carried out. The results show that seven groups of data are analyzed under the consideration of calculation time and accuracy, and the image resolution is set to 0.3-0.4 渭 m. The single cell model was irradiated with 200 偽 particles of 3MeV, and the average specific energy of a single particle in the nucleus was 0.209 Gy. The average specific energy of a single particle in the cytoplasm was 0.044 GY. The results obtained in this paper are in agreement with the published data. The method of establishing a single cell voxel model is feasible and universal. Nuclear density and cell morphology have important influence on the results of the model. It is necessary to establish cell banks of different cell lines for the study of radiobiology.
