【摘要】：目的觀察補(bǔ)體片段C3f對人胚肺成纖維細(xì)胞(MRC-5)合成α-平滑肌肌動蛋白(α-SMA)的影響。方法將低中高3個不同質(zhì)量濃度(15.6,62.5,250 ng·mL-1)人工合成的補(bǔ)體片段C3f(實(shí)驗(yàn)組)加入培養(yǎng)基中,以刺激MRC-5細(xì)胞,以細(xì)胞免疫組化法檢測α-SMA在MRC-5細(xì)胞質(zhì)中的表達(dá)情況。同時,設(shè)立對照組:10 ng·mL~(-1)轉(zhuǎn)化生長因子-β_1(TGF-β_1)和空白組(濃度為0的C3f),進(jìn)行對比研究。結(jié)果中劑量實(shí)驗(yàn)組與對照組(TGF-β_1)刺激MRC-5細(xì)胞合成α-SMA量分別為7.46±0.66,7.54±0.51,差異無統(tǒng)計(jì)學(xué)意義(P0.05),說明2組的量相當(dāng),表明C3f具有刺激MRC-5合成α-SMA的功能。低中高3個不同質(zhì)量濃度的細(xì)胞內(nèi)α-SMA表達(dá)量分別為4.95±0.78,7.46±0.66和9.23±0.21,顯示C3f的濃度與α-SMA的表達(dá)量呈正相關(guān)(P0.05),表明C3f具有促進(jìn)MRC-5表達(dá)α-SMA的功能。結(jié)論補(bǔ)體片段C3f能夠刺激MRC-5細(xì)胞內(nèi)α-SMA的合成增加。
[Abstract]:Objective to observe the effect of complement fragment C 3f on the synthesis of 偽 -smooth muscle actin (偽-SMA) in human embryonic lung fibroblasts (MRC-5). Methods three synthetic complement fragments C3f (experimental group) of low, medium and high concentration (15.6n 62.5250 ng mL-1) were added to the medium to stimulate MRC-5 cells, and the expression of 偽 SMA in the cytoplasm of MRC-5 was detected by immunohistochemical method. At the same time, 10 ng mL~ (-1) transforming growth factor- 尾 _ 1 (TGF- 尾 _ 1) and control group (C _ 3f with concentration of 0) were set up for comparative study. Results the amount of 偽-SMA stimulated by TGF- 尾 _ 1 in the middle dose group and the control group was 7.46 鹵0.666 鹵7.54 鹵0.51, respectively. There was no significant difference between the two groups (P0.05), indicating that C3f had the function of stimulating MRC-5 to synthesize 偽 -SMA. The expression levels of 偽-SMA were 4.95 鹵0.78 鹵7.46 鹵0.66 and 9.23 鹵0.21, respectively, indicating that C3f was positively correlated with the expression of 偽-SMA (P0.05), indicating that C3f could promote the expression of 偽-SMA by MRC-5. Conclusion C3f can stimulate the synthesis of 偽-SMA in MRC-5 cells.
