骨髓間充質(zhì)干細(xì)胞移植對(duì)矽肺大鼠肺纖維化的作用

【摘要】:目的 探討骨髓間充質(zhì)干細(xì)胞(BMSCs)移植對(duì)矽肺大鼠肺組織纖維化的影響。方法 將42只SD雌性大鼠分為A、B、C、D組。A、B和C組采用暴露式氣管內(nèi)一次性緩慢注入二氧化硅(SiO2)混懸液建立矽肺大鼠模型,D組則注入生理鹽水。B、C組分別于造模后6h或第42天經(jīng)尾靜脈注射BMSCs。第14天,A、B和D組分別處死大鼠各6只,第56天各組分別處死6只。HE染色觀察肺組織形態(tài)學(xué)變化,ELISA檢測肺組織羥脯氨酸(Hyp)水平。結(jié)果 A組在造模后第14天,肺組織病理改變以急性炎癥反應(yīng)為主,第56天以纖維化為主;B、C組病理改變均較同時(shí)期A組輕;D組未見明顯異常。肺組織中Hyp水平,A、B和C組均較D組明顯升高(P=0.000);B、C組與A組比較,在BMSCs治療后第14天均明顯減低(P=0.000、0.035),B組在治療后第56天仍明顯減低(P=0.016)。結(jié)論 不論在以肺泡炎為主還是在以纖維化為主的病理階段予BMSCs移植,均可以延緩矽肺大鼠肺纖維化的病理進(jìn)程。
[Abstract]:Objective to investigate the effect of bone marrow mesenchymal stem cell (BMSCs) transplantation on pulmonary fibrosis in silicosis rats. Methods Forty-two female SD rats were divided into two groups: group A (B) and group C (group C) were injected with exposed intratracheal silica (SiO2) suspension, and group D (group D) were injected with normal saline, respectively, 6 hours or the first time after the establishment of the model. 42 days BMSCs. via caudal vein On the 14th day, 6 rats were killed in group A B and 6 rats in group D, 6 rats in each group were killed on the 56th day. Morphologic changes of lung tissue were observed by HE staining and hydroxyproline (Hyp) level of lung tissue was detected by ELISA. Results the pathological changes of lung tissue in group A were mainly acute inflammatory reaction and fibrosis on the 14th day after modeling, the pathological changes in group B C were lighter than those in group A at the same time, and no obvious abnormality was found in group D. The level of Hyp in lung tissue in group A B and C was significantly higher than that in group D (P0. 000), and that in group C was significantly lower than that in group A on the 14th day after treatment (P < 0. 0000. 035), B) (P0. 016). Conclusion BMSCs transplantation can delay the pathological process of pulmonary fibrosis in silicosis rats.
