萊菔硫烷對(duì)染鎘小鼠睪丸間質(zhì)細(xì)胞抗氧化功能影響的研究

【摘要】:試驗(yàn)旨在研究萊菔硫烷(SFN)對(duì)染鎘(Cd)小鼠睪丸間質(zhì)細(xì)胞(TM3細(xì)胞)抗氧化應(yīng)激能力的影響。向TM3細(xì)胞培養(yǎng)液中添加Cd和SFN,分別測(cè)定Cd的半數(shù)抑制濃度(IC50)和SFN的安全劑量范圍,建立試驗(yàn)?zāi)P?通過(guò)檢測(cè)TM3細(xì)胞的相對(duì)存活率、乳酸脫氫酶(LDH)活性及細(xì)胞抗氧化水平,判定SFN對(duì)Cd誘導(dǎo)TM3細(xì)胞毒性的影響。結(jié)果顯示:1隨著Cd濃度的增加,TM3細(xì)胞的相對(duì)存活率降低,Cd對(duì)體外培養(yǎng)TM3細(xì)胞的IC50為51.4μmol/L;SFN在一定范圍內(nèi)對(duì)細(xì)胞存活有促進(jìn)作用,但超過(guò)范圍后具有毒性,且濃度越大毒性越大,本試驗(yàn)條件下選擇SFN濃度為2.5、5和10μmol/L。2與對(duì)照組相比,Cd組TM3細(xì)胞的T-SOD、GSH-Px活性及GSH含量顯著或極顯著降低(P0.05;P0.01),LDH活性、MDA含量升高;SFN組LDH活性、MDA含量降低,GSH含量、T-SOD活性及GSH-Px活力升高;與Cd組相比,Cd+SFN組GSH含量、T-SOD活性及GSH-Px活力顯著或極顯著升高(P0.05;P0.01);LDH活性、MDA含量則降低,表明SFN具有緩解染Cd誘導(dǎo)TM3細(xì)胞毒性的作用。
[Abstract]:The aim of this study was to investigate the effect of sulforaphane (SFN) on antioxidant stress of testis interstitial cells (TM3 cells) in cadmium-exposed (Cd) mice. The half inhibitory concentration (IC50) of Cd and the safe dose range of SFN were measured by adding Cd and SFN, to the culture medium of TM3 cells, and the experimental model was established. The relative survival rate of TM3 cells, the activity of lactate dehydrogenase (LDH) and the antioxidant level of TM3 cells were measured. To evaluate the effect of SFN on TM3 cell toxicity induced by Cd. The results showed that: 1 with the increase of Cd concentration, the relative survival rate of TM3 cells decreased. Cd promoted the survival of TM3 cells in a range of 51.4 渭 mol/L;SFN, but beyond the range was toxic, and the greater the concentration was, the greater the toxicity was. Compared with the control group, the activity of T-SODD-GSH-Px and the content of GSH in TM3 cells of Cd group were significantly or extremely decreased (P0.05P0.01), LDH activity, MDA content increased, LDH activity, MDA content decreased, GSH content, T-SOD activity and GSH-Px activity increased in SFN group compared with the control group, the concentration of SFN was 2.5 渭 mol/L.2 and 10 渭 mol/L.2, respectively). Compared with Cd group, GSH content, T-SOD activity and GSH-Px activity in, Cd SFN group were significantly increased (P0.05 + P0.01); LDH activity, MDA content was decreased, indicating that SFN could alleviate TM3 cytotoxicity induced by Cd.
