WSC-DTPA納米粒輻射防護(hù)作用的初步研究

【摘要】:為研究WSC-DTPA(水溶性低分子量殼聚糖WSC,二乙烯三胺五乙酸DTPA)納米粒的輻射防護(hù)作用,采用N-乙酰化反應(yīng)和離子凝膠法制備不同游離氨基含量的WSC-DTPA納米粒;MTT法檢測(cè)其對(duì)6 Gy60Coγ射線照射后48 h BRL細(xì)胞存活率的影響;活細(xì)胞工作站觀察BRL細(xì)胞攝取FITC-WSC-DTPA納米熒光探針的情況。結(jié)果表明:成功合成了游離氨基含量分別為92.7%、74.3%、1.59%的WSC-DTPA聚合物;WSC、WSC納米粒以及WSC-DTPA納米粒(氨基含量為92.7%,濃度在6.25μg/mL以上),隨著藥物濃度的增加,BRL細(xì)胞存活率均顯著高于單純照射組,差別有統(tǒng)計(jì)學(xué)意義(p0.05),而游離氨基含量為1.59%的WSC-DTPA納米粒無(wú)輻射保護(hù)作用;活細(xì)胞工作站檢驗(yàn)結(jié)果顯示2 h內(nèi)WSC納米粒、WSC-DTPA納米粒能夠進(jìn)入BRL細(xì)胞,而非納米化的WSC-DTPA聚合物無(wú)法進(jìn)入細(xì)胞。
[Abstract]:In order to study the radiation protection of WSC-DTPA (water-soluble low molecular weight chitosan WSC, diethylenetriamine pentaacetic acid DTPA) nanoparticles, WSC-DTPA nanoparticles with different free amino groups were prepared by N-acetylation reaction and ionic gel method. The effects of WSC-DTPA nanoparticles on the survival rate of BRL cells 48 h after irradiation with 6 Gy60Co 緯 -rays were measured. Live cell workstation was used to observe the uptake of FITC-WSC-DTPA nanofluorescent probe by BRL cells. The results showed that the WSC-DTPA polymer WSC nanoparticles and WSC-DTPA nanoparticles with a free amino group of 92.74.3and 1.59% were successfully synthesized (the amino content was 92.7and the concentration was above 6.25 渭 g/mL). With the increase of the drug concentration, the survival rate of WSC-DTPA cells was significantly higher than that of the irradiation group. The difference was statistically significant (p0.05), but the free amino content of 1.59% WSC-DTPA nanoparticles had no radiation protection. The results of living cell workstation test showed that WSC nanoparticles WSC-DTPA nanoparticles could enter BRL cells within 2 hours. The non-nanocrystalline WSC-DTPA polymer does not enter the cell.
