神經(jīng)毒性體外評價系統(tǒng)研究進展

【摘要】:神經(jīng)毒性體外評價系統(tǒng)是以細胞培養(yǎng)、組織培養(yǎng)等實驗方法為基礎(chǔ),利用生物化學(xué)、分子學(xué)、電生理學(xué)、形態(tài)學(xué)檢驗以及各種組學(xué)技術(shù)對毒物暴露致神經(jīng)系統(tǒng)結(jié)構(gòu)或功能損傷進行研究的評價體系。應(yīng)用領(lǐng)域包括高通量篩選神經(jīng)毒性化學(xué)藥物和神經(jīng)毒性作用機制的研究。體外評價系統(tǒng)與標(biāo)準(zhǔn)化的體內(nèi)神經(jīng)毒性實驗相比,其優(yōu)勢在于靈敏度高、可重復(fù)性強,是更加有效和經(jīng)濟的替代方法,能夠補充或增加體內(nèi)實驗數(shù)據(jù)。雖然體外評價系統(tǒng)具有諸多優(yōu)點,但也不能完全替代更加復(fù)雜和完整的神經(jīng)系統(tǒng)。如何將體外實驗數(shù)據(jù)與體內(nèi)實驗數(shù)據(jù)相關(guān)聯(lián)是研究者關(guān)注的主要領(lǐng)域。體外評價系統(tǒng)的研究不斷發(fā)展,如細化檢測終點、構(gòu)建特定的組織培養(yǎng)模型以及拓展簡化系統(tǒng)實現(xiàn)跨學(xué)科間的創(chuàng)新等。神經(jīng)毒性體外評價系統(tǒng)不僅僅是風(fēng)險性評價的主要方法,還是研究多類型神經(jīng)毒物作用機制的重要方法。
[Abstract]:Neurotoxicity evaluation system in vitro is based on cell culture, tissue culture and other experimental methods, using biochemical, molecular, electrophysiology, An evaluation system for structural or functional damage to the nervous system caused by exposure to toxic substances by morphological examination and various techniques. Applications include high-throughput screening of neurotoxic chemicals and mechanisms of neurotoxicity. Compared with the standard in vivo neurotoxicity test, the in vitro evaluation system has the advantages of high sensitivity, strong repeatability, and is a more effective and economical alternative method, which can supplement or increase in vivo experimental data. Although in vitro evaluation system has many advantages, it can not completely replace the more complex and complete nervous system. How to correlate in vitro experimental data with in vivo experimental data is a major area of concern for researchers. The research of in vitro evaluation system has been developing continuously, such as refinement of detection endpoint, construction of specific tissue culture model and expansion of simplified system to realize interdisciplinary innovation. Neurotoxicity in vitro evaluation system is not only the main method of risk assessment, but also an important method to study the mechanism of neurotoxicity.
