多不飽和脂肪酸飲食對載脂蛋白E缺失小鼠含載脂蛋白B100脂蛋白代謝的影響

【摘要】:探討不同多不飽和脂肪酸飲食對載脂蛋白E(apoE)缺失小鼠含apoB100脂蛋白代謝的影響.2月齡雄性載脂蛋白E缺失小鼠添加2%紫蘇籽油或紅花油飲食干預(yù)6周,檢測其血脂和血清apoB100水平;測定肝臟組織中HMG-Co A還原酶活性;使用實(shí)時定量PCR分析肝組織中含apoB100脂蛋白代謝相關(guān)基因表達(dá);蛋白質(zhì)印跡(Western blot)檢測PPARα蛋白表達(dá);油紅O染色檢測肝細(xì)胞中脂質(zhì)積累.結(jié)果顯示,紫蘇籽油飲食干預(yù)可降低血清LDL-C和apoB100水平,降低肝組織中HMG-Co A還原酶活性以及apoB100、FAS、HMG-Co AR、ACAT2、LRP、LDLR和SREBP-1c基因mRNA的表達(dá),顯著升高轉(zhuǎn)錄因子PPARα的mRNA和蛋白質(zhì)水平,明顯降低肝臟中脂質(zhì)沉積.而紅花油飲食干預(yù)后升高了小鼠血清甘油三酯和高密度脂蛋白膽固醇以及肝臟apoB100的mRNA表達(dá)水平,降低PPARα的mRNA和蛋白質(zhì)水平,明顯促進(jìn)肝臟中脂質(zhì)積累.結(jié)果提示在飲食中添加紫蘇籽油可有效降低含apoB100脂蛋白的代謝,對動脈粥樣硬化以及脂肪肝等疾病有更好的保護(hù)作用,對于正常人群以及家族性Ⅲ型高脂蛋白血癥患者的日常膳食提供理論依據(jù).
[Abstract]:Effects of different polyunsaturated fatty acid diets on lipoprotein metabolism in apolipoprotein E (apoE) deficient mice the effects of 2% perilla seed oil or safflower oil on the metabolism of apoB100 lipoprotein in 2% perilla seed oil or safflower oil in male apolipoprotein E deficient mice were studied for 6 weeks. The activity of HMG-Co A reductase in liver tissue was measured, the expression of apoB100 lipoprotein related genes in liver tissue was analyzed by real-time quantitative PCR, the expression of PPAR 偽 protein was detected by Western blot (Western blot), and the lipid accumulation in hepatocytes was detected by oil red O staining. The results showed that the dietary intervention of perilla seed oil could decrease the levels of serum LDL-C and apoB100, decrease the activity of HMG-Co A reductase and the expression of mRNA and mRNA of PPAR 偽. The lipid deposition in the liver was significantly decreased. The levels of serum triglyceride and high density lipoprotein cholesterol (HDL-C) and mRNA expression of apoB100 in liver were increased, mRNA and protein levels of PPAR 偽 were decreased, and lipids accumulation in liver was significantly increased after the intervention of safflower oil diet. The results suggest that the addition of perilla seed oil to diet can effectively reduce the metabolism of apoB100 lipoprotein and has a better protective effect on atherosclerosis and fatty liver.
