【摘要】：綜述了在21世紀(jì)毒性測(cè)試愿景與策略倡議下,以及現(xiàn)代科技發(fā)展和法規(guī)監(jiān)管體系變革的背景下,有害結(jié)局通路(AOP)的提出和發(fā)展。重點(diǎn)介紹了AOP的概念和指南,AOP的基本組成、構(gòu)建條件和信息共享,AOP的網(wǎng)絡(luò)和評(píng)估;討論了AOP在皮膚致敏和魚類早期發(fā)育2個(gè)毒性終點(diǎn)的開發(fā)和應(yīng)用情況,及其作為自上而下和從中間開始的2種AOP開發(fā)策略的實(shí)例;最后提出了AOP以現(xiàn)代科技發(fā)展為動(dòng)力、毒性預(yù)測(cè)現(xiàn)代化為基礎(chǔ)、推動(dòng)法規(guī)監(jiān)管模式變革為目標(biāo)的發(fā)展策略。AOP指南也為替代動(dòng)物測(cè)試方法的開發(fā)和檢測(cè)技術(shù)創(chuàng)新提供了難得的機(jī)遇。
[Abstract]:Under the initiative of vision and strategy of toxicity testing in the 21st century and the background of the development of modern science and technology and the reform of regulatory system, the harmful outcome pathway (AOPs) was proposed and developed. The concept of AOP and the basic composition of AOP, the construction conditions and the network and evaluation of AOP for information sharing are introduced, and the development and application of AOP in skin sensitization and fish early development are discussed. As an example of two AOP development strategies from top to bottom and from the middle, it is proposed that AOP is based on the development of modern science and technology and the modernization of toxicity prediction. AOP guidelines also provide a rare opportunity for the development of alternative animal testing methods and the innovation of testing techniques.
