【摘要】：目的研究吸入高濃度氫氣后對四氯化碳損傷性大鼠生精細胞內(nèi)Akt表達的影響。方法選取健康雄性3月齡SD大鼠30只,隨機平均分為對照組(Con組),四氯化碳組(CCl_4組)和氫氣組(H_2組)。Con組大鼠于每周一、周五皮下注射橄欖油(0.07ml/100g),CCl_4組和H_2組于每周一、周五皮下注射30%CCl_4+70%橄欖油(0.1ml/100g),持續(xù)6周;H_2組在第6周加入高濃度H_2(3%H_2+97%空氣)治療,每天1h,連續(xù)7d。取睪丸,應(yīng)用免疫組織化學(xué)和Western blot方法檢測Akt的表達水平。結(jié)果免疫組化結(jié)果顯示,Akt主要表達于精原細胞和初級精母細胞的胞漿中,CCl_4組大鼠生精細胞內(nèi)Akt表達水平較對照組明顯降低,H_2組較CCl_4組表達水平升高(P0.01)。Western blot結(jié)果顯示與免疫組化結(jié)果相一致。結(jié)論高濃度H_2可以增加CCl_4損傷性大鼠睪丸組織中Akt的表達。
[Abstract]:Objective to study the effect of inhaling high concentration of hydrogen on the expression of Akt in the spermatogenic cells of rats with carbon tetrachloride injury. Methods 30 healthy male 3 month old SD rats were randomly divided into control group (group Con), carbon tetrachloride group (group CCl_4) and hydrogen group (H_2 group).Con rats were subcutaneously injected with olive oil (0.07ml/100g) and CCl_4 on Friday (0.07ml/100g). Group and H_2 group were subcutaneously injected with 30%CCl_4+70% olive oil (0.1ml/100g) every Monday and Friday for 6 weeks. Group H_2 was treated with high concentration of H_2 (3%H_2+97% air) at sixth weeks, 1H, 7d. taking every day, immunohistochemistry and Western blot method were used to detect Akt expression. Results immunohistochemistry showed that Akt was mainly expressed in spermatogonia. In the cytoplasm of cells and primary spermatocytes, the expression level of Akt in the spermatogenic cells of group CCl_4 was significantly lower than that in the control group. The expression level of.Western blot in group H_2 was higher than that in CCl_4 group (P0.01), and the results of.Western blot showed the same with the immunohistochemical results. Conclusion high concentration H_2 can increase the expression of Akt in the testicular tissue of rats with CCl_4 injury.
