母源性丙烯酰胺暴露對斷乳期大鼠額葉皮質(zhì)GAP-43和BDNF表達的影響

【摘要】:目的:探討母源性丙烯酰胺(acrylamide,ACR)暴露對斷乳期大鼠額葉皮質(zhì)生長相關(guān)蛋白(GAP-43)和腦源性神經(jīng)營養(yǎng)因子(BDNF)表達的影響。方法:SD孕鼠隨機分為4組,懷孕第6 d起對照組和實驗組分別給予雙蒸水和50,100,200μg/ml ACR飲水,直至仔鼠出生21 d,染毒結(jié)束時對斷乳期大鼠稱重。免疫組織化學(xué)檢測額葉皮質(zhì)GAP-43和BDNF的表達。結(jié)果:與對照組相比,實驗各組斷乳期大鼠體重呈濃度依賴性下降,且組間差異明顯(P0.05)。與對照組相比,ACR實驗各組GAP-43表達顯著降低(P0.05),而低、中劑量組之間差異不顯著(P0.05),高劑量組與低、中劑量組相比,表達顯著減少(P0.05);與對照組相比,ACR實驗組各組BDNF表達顯著減少(P0.05),中、高劑量組與低劑量組相比,BDNF表達減少(P0.05),而中、高劑量組之間差異不顯著(P0.05)。結(jié)論:母源性ACR染毒可能通過下調(diào)額葉皮質(zhì)神經(jīng)元GAP-43和BD)NF表達,抑制軸突延伸或擾亂神經(jīng)元發(fā)育的內(nèi)環(huán)境穩(wěn)態(tài),進而影響額葉皮質(zhì)的發(fā)育。
[Abstract]:Aim: to investigate the effects of maternal acrylamide (ACR) exposure on the expression of GAP-43 in frontal cortex and BDNF in rat frontal cortex. Methods Twenty SD pregnant rats were randomly divided into 4 groups. From the 6th day after pregnancy, the control group and the experimental group were given double steamed water and 50100200 渭 g/ml ACR drinking water, respectively, until 21 days after birth, and the weaning rats were weighed at the end of the exposure. The expression of GAP-43 and BDNF in frontal cortex was detected by immunohistochemistry. Results: compared with the control group, the body weight of the weaning rats decreased in a concentration-dependent manner, and the difference between the two groups was significant (P 0.05). Compared with the control group, the expression of GAP-43 in the ACR experimental group was significantly lower than that in the control group, but in the middle dose group there was no significant difference between the two groups. The expression of BDNF in the high dose group was significantly lower than that in the low group and in the middle dose group, and the expression of BDNF in the control group was significantly lower than that in the control group, and the expression of BDNF in the experimental group was significantly lower than that in the control group. The expression of BDNF in high dose group was lower than that in low dose group, but there was no significant difference between high dose group and high dose group. Conclusion: maternal ACR exposure may inhibit the extension of axons or disturb the homeostasis of neurons by down-regulating the expression of GAP-43 and BD)NF in the frontal cortex and thus affect the development of the frontal cortex.
