【摘要】：為了評估金桔的食用風(fēng)險,試驗采用基質(zhì)輔助激光解吸電離飛行時間質(zhì)譜法(MALDI-TOF-MS)調(diào)查金桔表面腸桿菌科微生物分布情況,篩選致病微生物,同時采用與生化鑒定和16S rDNA測序鑒定輔助鑒定結(jié)果。實驗共分離6種的腸桿菌科細(xì)菌,分別為日溝維腸桿菌、分散泛菌、肺炎克雷伯菌、阪崎腸桿菌、生癌腸桿菌、陰溝腸桿菌,其中分散泛菌、陰溝腸桿菌和肺炎克雷伯菌出現(xiàn)頻率較高;對三種鑒定方法進行比較分析,MALDI-TOF-MS鑒定結(jié)果與另外兩種與屬水平一致,種水平有偏離;MALDI-TOF-MS對分離微生物進行聚類分析表明同屬微生物親緣較近,同種微生物峰譜圖仍有差異。試驗結(jié)果表明,金桔表面具有腸道致病微生物,有一定食用風(fēng)險,建議加強農(nóng)產(chǎn)品致病菌常規(guī)的檢測及風(fēng)險評估力度;MALDI-TOF-MS方法可分辨率高且能提供基于蛋白質(zhì)水平歸類分析可以用于農(nóng)產(chǎn)品微生物的快速檢測。
[Abstract]:In order to evaluate the edible risk of kumquat, MALDI-TOF-MSMS was used to investigate the distribution of Enterobacteriaceae on the surface of Citrus kumquat, and to screen the pathogenic microbes. At the same time, the results were identified with biochemical identification and 16s rDNA sequencing. A total of 6 species of Enterobacteriaceae were isolated. They were Enterobacter hirsurea, pantobacillus dispersus, Klebsiella pneumoniae, Enterobacter sakazakii, Enterobacter carcinogenes, Enterobacter cloacae, among which pan-bacteria were dispersed. The frequencies of Enterobacter cloacae and Klebsiella pneumoniae were high, and the results of MALDI-TOF-MS identification were consistent with those of the other two. The cluster analysis of isolated microorganisms by MALDI-TOF-MS showed that the phylogeny of the same species was close and the peak spectrum of homologous microbes was still different. The results showed that there were intestinal pathogenic microorganisms on the surface of kumquat, and there was a certain edible risk. It is suggested that the conventional detection and risk assessment of pathogenic bacteria in agricultural products should be strengthened. MALDI-TOF-MS can provide high resolution and can be used for rapid detection of agricultural microbes based on protein level classification analysis.
