N-氨基甲酰-L-谷氨酸對斷奶小鼠生長性能的影響

【摘要】:動(dòng)物由于斷奶應(yīng)激會出現(xiàn)無食欲、免疫力下降、生長減緩等多種癥狀,研究證明斷奶后由于飼料中精氨酸(Arg)不能滿足動(dòng)物生長的需要,是這些癥狀形成的重要原因之一。因此提高動(dòng)物斷奶后內(nèi)源性精氨酸的合成可以有效減輕斷奶應(yīng)激反應(yīng),更好的促進(jìn)斷奶動(dòng)物的生長。 N-氨甲酰-L-谷氨酸(N-Carbamyl-L-glutamic acid, NCG)是N-乙酰-L-谷氨酸(N-Acetyl-L-glutamic acid, NAG)的化學(xué)結(jié)構(gòu)類似物。在動(dòng)物體內(nèi),它能產(chǎn)生與NAG相同的生理功能——催化氨甲酰磷酸合成酶-I(CPS-I)的生成,進(jìn)而促進(jìn)谷氨酰胺、脯氨酸等轉(zhuǎn)化為精氨酸,從而增加精氨酸的內(nèi)源性合成,另外由于細(xì)胞內(nèi)沒有降解NCG的脫酰酶,NCG幾乎不被分解,因而很小的劑量就能產(chǎn)生顯著作用。因此,NCG對改善動(dòng)物斷奶后精氨酸合成及吸收量不能滿足生長代謝需要而引起的各種癥狀會產(chǎn)生較好的效果。本文對NCG及其與Arg的聯(lián)合作用對斷奶小鼠生長性能、血清生化指標(biāo)、血清精氨酸含量、免疫器官指數(shù)的影響進(jìn)行了研究。 主要研究內(nèi)容與結(jié)果歸納如下: 1.研究了日糧中添加不同濃度NCG對斷奶小鼠生長性能的影響。在斷奶小鼠基礎(chǔ)日糧中添加0.025%、0.05%、0.1%的NCG,通過監(jiān)測小鼠體重的變化,平均日增重、日采食量、料重比等數(shù)據(jù)研究NCG對斷奶小鼠生長性能的影響。實(shí)驗(yàn)結(jié)果發(fā)現(xiàn)日糧中添加0.1%的NCG,斷奶小鼠平均日增質(zhì)量與對照組相比提高了12.40%,飼料與體質(zhì)量比與對照組相比下降了14.65%。結(jié)果表明日糧中添加適量濃度的NCG能顯著提高斷奶小鼠生長性能。 2.研究了NCG與Arg聯(lián)合作用對斷奶小鼠生長性能的影響。選擇在基礎(chǔ)日糧中同時(shí)添加0.05%的NCG和三個(gè)水平的Arg(0.025%、0.05%、0.1%),研究NCG與Arg聯(lián)合作用對斷奶小鼠生長性能的影響。實(shí)驗(yàn)結(jié)果發(fā)現(xiàn)同時(shí)添加0.05%的NCG和0.025%的Arg,斷奶小鼠平均日增質(zhì)量提高了10.53%。結(jié)果表明適量濃度NCG與Arg的聯(lián)合作用,能顯著提高斷奶小鼠生長性能,并且提高生長性能的效果比單獨(dú)添加NCG、Arg要好。同時(shí)還發(fā)現(xiàn)單獨(dú)添加Arg只能在斷奶后初始階段對生長性能有較好的促進(jìn)作用,之后效果不佳,而0.1%NCG組和低劑量Arg聯(lián)合作用在整過實(shí)驗(yàn)階段都有較好的促生長作用。 3.研究了NCG對斷奶小鼠血清中谷丙轉(zhuǎn)氨酶(GPT)、血清尿素氮(BUN)、總蛋白、白蛋白、球蛋白含量的影響。結(jié)果表明NCG及其與Arg聯(lián)合作用對斷奶小鼠血清GPT沒有顯著影響;高NCG組和高Arg聯(lián)合組BUN值顯著高于對照組,推測高濃度NCG及高濃度Arg與中劑量NCG聯(lián)合作用可能對小鼠腎臟造成一定的損傷。高NCG組、NCG與Arg聯(lián)合作用組與對照組相比總蛋白和白蛋白都沒有顯著差異,中NCG組總蛋白、白蛋白含量顯著低于對照組,說明NCG促進(jìn)生長性能的提高與血清中總蛋白、白蛋白的含量沒有直接聯(lián)系。 3.研究了NCG對斷奶小鼠血清中游離Arg的質(zhì)量濃度的影響。實(shí)驗(yàn)結(jié)果發(fā)現(xiàn),在基礎(chǔ)日糧中添加0.05%和0.1%的NCG,都能顯著提高血清中游離Arg的質(zhì)量濃度,與對照組相比分別提高了21.00%和42.77%,并且數(shù)據(jù)表明NCG引起機(jī)體內(nèi)Arg合成量的變化并不與NCG的添加量成正比。綜合平均日增質(zhì)量和血清游離Arg含量數(shù)據(jù)發(fā)現(xiàn),生長速率的提高與提高機(jī)體內(nèi)Arg的轉(zhuǎn)化量有關(guān)系但不成正比例,推測NCG在提高生長性能的貢獻(xiàn)上除了提高內(nèi)源性Arg合成量外還有其它的作用。 4.研究了NCG對斷奶小鼠免疫器官指數(shù)的影響。實(shí)驗(yàn)發(fā)現(xiàn)NCG對斷奶小鼠脾臟指數(shù)的影響有一定的線性關(guān)系,但作用不顯著,對胸腺指數(shù)影響較小。NCG與Arg聯(lián)合作用組斷奶小鼠脾臟指數(shù)和胸腺指數(shù)與空白對照組相比都沒有顯著差異,表明NCG與Arg聯(lián)合作用對斷奶以后17天小鼠免疫器官指數(shù)沒有顯著性作用。
[Abstract]:It is one of the important reasons for the formation of these symptoms after weaning because of the inability of arginine (Arg) in feed to meet the needs of animal growth. Therefore, the improvement of endogenous arginine synthesis can effectively reduce weaning stress after weaning. The reaction is better to promote the growth of the weaned animals.
N- -L- glutamic acid (N-Carbamyl-L-glutamic acid, NCG) is a chemical structural analogue of N- acetyl -L- glutamic acid (N-Acetyl-L-glutamic acid, NAG). In animals, it can produce the same physiological function as NAG, catalyze the formation of aminoformyl phosphate synthetase -I, and then promote the transformation of glutamine, proline, and so on into arginine. Acid, thus increasing the endogenous synthesis of arginine, and in addition to the lack of dedeoylase of NCG in cells, NCG is almost not decomposed, so a very small dose can produce significant effects. Therefore, NCG has a good effect on the various symptoms caused by the improvement of the synthesis and absorption of arginine after weaning in animals that can not meet the needs of growth metabolites. The effects of the combination of NCG and Arg on the growth performance, serum biochemical indexes, serum arginine content and immune organ index of weaned mice were studied in this paper.
The main research contents and results are summarized as follows:
1. the effects of adding different concentrations of NCG on the growth performance of weaned mice were studied. Adding 0.025%, 0.05%, 0.1% NCG in the basal diet of weaned mice, the effects of NCG on the growth performance of weaned mice were studied by monitoring the changes of weight of mice, average daily weight gain, daily feed intake and feed weight ratio. The experimental results showed that the diet added 0 to the diet. .1% NCG, the average daily mass of weaning mice increased by 12.40% compared with the control group. The feed and body mass ratio decreased significantly compared with the control group. The addition of a proper concentration of NCG in the 14.65%. result table could significantly increase the growth performance of the weaned mice.
2. the effect of the combination of NCG and Arg on the growth performance of weaned mice was studied. The effects of the combination of 0.05% NCG and three levels of Arg (0.025%, 0.05%, 0.1%) on the basal diet were selected to study the effects of the combined action of NCG and Arg on the growth performance of weaned mice. The experimental results showed that 0.05% NCG and 0.025% Arg were added to the experiment, and the average day of weaning mice was found. The 10.53%. results showed that the combined effect of appropriate concentration of NCG and Arg could significantly improve the growth performance of weaned mice, and the effect of increasing growth performance was better than that of adding NCG alone and Arg. At the same time, it was found that the addition of Arg alone could have a better effect on Growth in the initial stage of weaning. After that, the effect was not good, and 0 The combined action of.1%NCG group and low dose Arg had a good growth promoting effect in the whole experimental stage.
3. the effects of NCG on alanine transaminase (GPT), serum urea nitrogen (BUN), total protein, albumin and globulin content in the serum of weaned mice were studied. The results showed that the combination of NCG and Arg had no significant effect on serum GPT in weaned mice, and the BUN value of high NCG group and high Arg group was significantly higher than that of the control group, and the high concentration NCG and high concentration Arg were presumed to be Arg. The combined action of the medium dose NCG could cause certain damage to the kidney of mice. In the high NCG group, there was no significant difference in total protein and albumin in the combination group of NCG and Arg compared with the control group. The content of total protein and albumin in the NCG group was significantly lower than that of the control group. It showed that the increase of the growth energy of the NCG and the content of the total protein and albumin in the serum There is no direct connection.
3. the effect of NCG on the mass concentration of free Arg in the serum of weaned mice was studied. The results showed that adding 0.05% and 0.1% NCG in the basal diet could significantly increase the concentration of free Arg in the serum, which was increased by 21% and 42.77% compared with the control group, and the data showed that the NCG caused the change of Arg synthesis in the body. It is directly proportional to the amount of NCG added. The comprehensive average daily mass and serum free Arg content data show that the increase of the growth rate is related to the increase of the transformation of Arg in the body, but it is not proportional. It is speculated that the contribution of NCG to the increase of growth performance is other than the increase of endogenous Arg synthesis.
4. the effect of NCG on the immune organ index of weanling mice was studied. It was found that NCG had a certain linear relationship with the spleen index of weaned mice, but the effect was not significant. There was no significant difference in spleen index and thymus index between.NCG and Arg in the group of weaning mice compared with that of the empty white control group, indicating that NCG Combined with Arg had no significant effect on immune organ index in 17 days after weaning.
