DIDP和DINP暴露對小鼠的免疫毒性作用

【摘要】:為了探究增塑劑鄰苯二甲酸二異壬酯(DINP)和鄰苯二甲酸二異癸酯(DIDP)聯(lián)合暴露對小鼠免疫系統(tǒng)毒性有無協(xié)同作用,實(shí)驗(yàn)采用56只SPF級(jí)雄性Balb/C小鼠,隨機(jī)分為7組,用同種濃度(200mg/kg)不同質(zhì)量配比的DIDP和DINP混合藥劑對小鼠進(jìn)行灌胃,染毒暴露周期為28d.染毒結(jié)束后,取耳組織,測定耳腫指數(shù)和雙耳重量差;取耳組織和血清,檢測生物因子IgE,IFN-γ,IL-4等.實(shí)驗(yàn)結(jié)果顯示,DIDP與DINP聯(lián)合口腔暴露使小鼠皮膚炎癥明顯加劇(P0.01),且隨著DINP質(zhì)量比例增大,免疫毒性逐漸增強(qiáng).這表明,DIDP和DINP聯(lián)合暴露,對小鼠有免疫系統(tǒng)毒性作用.但是,聯(lián)合暴露時(shí),各指標(biāo)均是呈遞增或遞減趨勢,并未出現(xiàn)以某種比例混合時(shí)加強(qiáng)或減弱免疫毒性作用的現(xiàn)象.這表明,DIDP和DINP聯(lián)合暴露時(shí)并未有拮抗或協(xié)同作用.
[Abstract]:In order to investigate the synergistic effect of plasticizer DINP (diisononyl phthalate) and diisodecyl phthalate (DIDP) on the immune system of mice, 56 male Balb/C mice of SPF grade were randomly divided into 7 groups. Mice were perfused with the mixture of DIDP and DINP of the same concentration of 200 mg / kg and the exposure period was 28 days. After the exposure, the ear tissue was removed, the ear swelling index and the weight difference of both ears were measured, and the biological factor IgE- IFN- 緯 IL-4 was detected in the ear tissue and serum. The results showed that the combined oral exposure of DIDP and DINP significantly aggravated the skin inflammation in mice, and the immunotoxicity increased with the increase of the mass ratio of DINP. This indicated that the combined exposure of DIDP and DINP had a toxic effect on the immune system of mice. However, when combined exposure, each index showed an increasing or decreasing trend, and there was no phenomenon of strengthening or weakening the immune toxicity when mixed in some proportion. This indicated that there was no antagonistic or synergistic effect between DIDP and DINP.
