【摘要】：目的本研究對(duì)線蟲(chóng)暴露于金納米團(tuán)簇和納米碳點(diǎn)及其偶聯(lián)抗體產(chǎn)物(共4種醫(yī)用納米探針)后的感知行為和表型指標(biāo)進(jìn)行初步研究,以期探索應(yīng)用線蟲(chóng)評(píng)價(jià)納米材料安全性的新模式。方法將模式生物線蟲(chóng)用于金納米團(tuán)簇和納米碳點(diǎn)的毒性研究,分別觀察納米材料對(duì)線蟲(chóng)的感知行為(趨避指數(shù)、趨向指數(shù))、線蟲(chóng)表型指征(體型、灰度值、卷曲度、活動(dòng)能力4個(gè)方面共16個(gè)指標(biāo))是否有改變,以判斷其毒性作用大小。結(jié)果金納米團(tuán)簇(≤3.00 mg/ml)、偶聯(lián)抗體金納米團(tuán)簇(≤3.00 mg/ml)、碳點(diǎn)(≤1.51 mg/ml)及偶聯(lián)抗體碳點(diǎn)(≤3.50 mg/ml)趨避實(shí)驗(yàn)為陰性;在趨向試驗(yàn)中,3.00、1.40和0.30 mg/ml金納米團(tuán)簇、0.33 mg/ml碳點(diǎn)的趨向指數(shù)為陽(yáng)性。16個(gè)非致死性指標(biāo)總陽(yáng)性率分別為偶聯(lián)抗體碳點(diǎn)(81%)偶聯(lián)抗體金納米團(tuán)簇(69%)金納米團(tuán)簇(56%)碳點(diǎn)(50%)。結(jié)論本研究初步探討了4種納米材料對(duì)線蟲(chóng)感知行為的影響,且將線蟲(chóng)的表型指征首次應(yīng)用于納米材料毒性研究中,為探索納米材料的毒性提供了新穎的、有一定參考價(jià)值的快速篩選的指標(biāo)。
[Abstract]:Objective to study the sensing behavior and phenotypic indexes of nematodes exposed to gold nanoclusters and carbon nanoparticles and their coupling antibody products (including four kinds of medical nanoprobes). In order to explore the application of nematodes to evaluate the safety of nanomaterials a new model.
