辛基酚對5-羥色胺轉(zhuǎn)運(yùn)及受體通路的影響

【摘要】:目的通過觀察人肝7702細(xì)胞暴露于辛基酚(OP)后5-羥色胺(5-HT)轉(zhuǎn)運(yùn)體SERT、5-HT3b受體表達(dá)以及5-HT水平的變化,探討OP對5-HT轉(zhuǎn)運(yùn)通路和受體結(jié)合通路的影響。方法實(shí)驗(yàn)分5個(gè)組,分別為OP高(5.0μg/L)、中(1.0μg/L)、低(0.2μg/L)劑量組、鹽酸氟西汀組和空白對照組,體外細(xì)胞培養(yǎng)48 h,采用ELISA試劑盒檢測細(xì)胞上清液中5-HT含量的變化,運(yùn)用實(shí)時(shí)熒光定量PCR法檢測轉(zhuǎn)運(yùn)體SERT及5-HT3b受體的表達(dá)。結(jié)果中、高劑量組細(xì)胞上清液中5-HT含量高于空白對照組(P0.05),細(xì)胞中5-HT3b受體的表達(dá)降低(P0.05);低、中、高劑量組細(xì)胞中SERT的表達(dá)均低于空白對照組(P0.05);與鹽酸氟西汀組檢測結(jié)果一致。結(jié)論 OP可引起5-HT再攝取障礙,同時(shí)減少5-HT3b受體的表達(dá),對機(jī)體5-HT代謝網(wǎng)絡(luò)的轉(zhuǎn)運(yùn)通路及受體結(jié)合通路產(chǎn)生毒效應(yīng)作用。
[Abstract]:Objective to investigate the effect of OP on 5-HT transport pathway and receptor binding pathway by observing the changes of 5- serotonin (5-HT) transporter SERT, 5-HT3b receptor expression and 5-HT level after exposure to octyl phenol (OP) in human liver 7702 cells. Methods 5 groups were divided into OP high (5 mu g/L), medium (1 mu g/L), low (0.2 micron) dose group, and fluoxetine hydrochloride group. In the blank control group, the cell culture was 48 h in vitro, and the change of 5-HT content in the cell supernatant was detected by ELISA kit. The expression of SERT and 5-HT3b receptor in the transporter was detected by real time fluorescence quantitative PCR method. The results showed that the content of 5-HT in the supernatant of high dose group was higher than that of the blank control group (P0.05), and the expression of 5-HT3b receptor in the cell was reduced (P0.05). The expression of SERT in the low, middle and high dose groups was lower than that in the blank control group (P0.05), which was consistent with the test results of the fluoxetine hydrochloride group. Conclusion OP can cause the 5-HT reuptake barrier and reduce the expression of 5-HT3b receptor, which has a toxic effect on the transport pathway of 5-HT metabolic network and the receptor junction pathway.
