槲皮素及其乙酸乙酯衍生物的平衡溶解度和油水分配系數(shù)的測定

【摘要】:通過測定槲皮素及其酯化物的平衡溶解度和油水分配系數(shù),檢測酯化后的槲皮素衍生物脂溶性是否提高。采用高效液相色譜法測定槲皮素及3,7,3’,4’-O-四乙酸乙酯槲皮素在水、不同緩沖鹽溶液中的質(zhì)量濃度;采用搖瓶-液相色譜法,測定二者在正辛醇-水、緩沖液體系中的油水分配系數(shù)。37℃時,槲皮素在水中的平衡溶解度為23.02μg/m L,而3,7,3’,4’-O-四乙酸乙酯槲皮素在水中的平衡溶解度更小,為10.23μg/m L。隨著溶劑p H值的增大,槲皮素的平衡溶解度增大,3,7,3’,4’-O-四乙酸乙酯槲皮素變化并不顯著。槲皮素、3,7,3’,4’-O-四乙酸乙酯槲皮素在正辛醇-水體系條件下的油水分配系數(shù)分別為1.819、3.696。酯化修飾后的3,7,3’,4’-O-四乙酸乙酯槲皮素脂溶性高于槲皮素。
[Abstract]:The equilibrium solubility and oil-water partition coefficient of quercetin and its esters were determined to determine whether the liposolubility of quercetin derivatives was increased after esterification.High performance liquid chromatography (HPLC) was used to determine the mass concentrations of quercetin and quercetin in water and different buffer salt solutions, and to determine the concentration of quercetin in n-octanol-water by shaking flask liquid chromatography.The equilibrium solubility of quercetin in water was 23.02 渭 g / mL at 37 鈩,
