【摘要】：目的探討氯化鎘(CdCl_2)對小鼠腦組織氧化損傷以及核因子κB(NF-κB)、腫瘤壞死因子-α(TNF-α)、可溶性小鼠細(xì)胞間粘附分子(ICAM-1)表達(dá)的影響。方法 32只健康成年ICR雄鼠隨機分為CdCl_2 2.0、1.0和0.5 mg/kg處理組及生理鹽水(NS)對照組,各組經(jīng)腹腔注射CdCl_2,連續(xù)14 d。末次染毒后進(jìn)行曠場實驗;頸椎脫臼處死取腦組織,測定活性氧(ROS)含量,ELISA法檢測TNF-α和ICAM-1的表達(dá),免疫組化及蛋白質(zhì)印跡法觀察腦組織NF-κB P65蛋白的表達(dá),HE染色大腦神經(jīng)病理學(xué)檢查。結(jié)果隨CdCl_2劑量的增加,實驗動物體重增長程度下降(P0.05),CdCl_2 2.0 mg/kg組體重增加程度顯著低于其他各組(P0.05)。在曠場實驗中,CdCl_2 2.0 mg/kg組跨越格子數(shù)顯著高于NS對照組(P0.05)。CdCl_2 2.0和1.0mg/kg組小鼠腦組織中ROS水平增高(P0.05)、CdCl_2 2.0 mg/kg組TNF-α表達(dá)增加(P0.05),ICAM-1各組間差異無統(tǒng)計學(xué)意義。免疫組化結(jié)果和Western blot均提示,CdCl_2 2.0 mg/kg組NF-κB P65表達(dá)增加。病理檢查發(fā)現(xiàn),CdCl_2 2.0 mg/kg組大腦皮質(zhì)及海馬層細(xì)胞稀疏。結(jié)論鎘暴露后小鼠腦組織中ROS表達(dá)增加,ROS可以激活NF-κB通路,上調(diào)炎性細(xì)胞因子TNF-α的表達(dá)。
[Abstract]:Objective to investigate the effects of cadmium chloride (CDCL _ 2) on oxidative injury and the expression of nuclear factor 魏 B (NF- B) NF- 魏 B, tumor necrosis factor- 偽 (TNF- 偽), soluble intercellular adhesion molecule (ICAM-1) in mice.Methods Thirty-two healthy adult male ICR rats were randomly divided into CdCl_2 2.0 mg/kg 1.0 and 0.5 mg/kg groups and normal saline control group. Each group was intraperitoneally injected with CDCL _ 2 for 14 days.After the last exposure, the open field test was carried out, and the brain tissue was killed and the reactive oxygen species (Ros) content was determined. The expression of TNF- 偽 and ICAM-1 was detected by Elisa.The expression of NF- 魏 B p65 protein in brain tissue was observed by immunohistochemistry and Western blotting.Results with the increase of CdCl_2 dose, the weight gain of the experimental animals decreased significantly in the group of P0.05CDCl22 mg/kg as compared with that in the other groups.In the open field experiment, the cross lattice number of CDCL 2. 0 mg/kg group was significantly higher than that of NS control group (P 0. 05). CDCL 2. 0. 0. There was no significant difference in the increase of TNF- 偽 expression in the brain tissue of the 1.0mg/kg group and the P0. 05% CDCL 2. 0 mg/kg group. There was no significant difference between the two groups in the expression of TNF- 偽 in the control group (P 0. 05).Immunohistochemical results and Western blot showed that NF- 魏 B p65 expression was increased in CDCL 2. 0 mg/kg group.Pathological examination showed that the cells in cortex and hippocampal layer were sparse in the group of 20 mg/kg CDCL 2. 0.Conclusion the increased expression of ROS in mouse brain after cadmium exposure can activate the NF- 魏 B pathway and up-regulate the expression of inflammatory cytokine TNF- 偽.
