蘋果葉總黃酮的提取及其含量測定

【摘要】:以"紅元帥"蘋果的干燥葉子為試材,通過單因素試驗確定蘋果葉總黃酮的最佳提取工藝條件,采用可見分光光度法對蘋果葉中總黃酮含量進行測定,以期為蘋果葉中總黃酮提取工藝及含量測定提供方法和依據。結果表明:蘋果葉總黃酮的最佳提取工藝條件為乙醇作溶劑,乙醇濃度60%,料液比1∶20g/mL,提取溫度為70℃,提取時間為30min;在選定試驗條件下對蘋果葉進行含量測定,對蘆丁對照品濃度與吸光度進行線性回歸,其回歸方程為Y=24.821 43X-0.031,R2=0.999 45(n=6),蘆丁對照品質量在4~24μg范圍內呈良好的線性關系,在此條件下測定蘋果葉中總黃酮平均含量為30.28mg/g。試驗優(yōu)選出了蘋果葉中總黃酮的最佳提取工藝條件及含量測定方法,該方法簡單、快速,準確,精密度,穩(wěn)定性,重現性良好。
