亮氨酸對高脂高膽固醇喂養(yǎng)小鼠瘦素抵抗影響

【摘要】:目的探討亮氨酸對高脂高膽固醇喂養(yǎng)小鼠體脂和瘦素抵抗的影響。方法 60只雄性C57BL/6J小鼠隨機分為對照組、高脂高膽固醇組、低、高劑量亮氨酸組(1.5%、3.0%亮氨酸),干預24周;測定小鼠體重、脂肪重量、血清膽固醇和瘦素水平,Western blot法檢測小鼠睪周脂肪組織中瘦素受體(Ob-R)、Janus蛋白酪氨酸激酶2(JAK2)、信號轉導和轉錄激活子3(STAT3)及細胞因子信號轉導抑制因子3(SOCS3)的蛋白表達。結果高脂高膽固醇喂養(yǎng)明顯升高小鼠體重、脂重和血清膽固醇水平;與高脂高膽固醇組比較,亮氨酸組小鼠體重、脂重和血清膽固醇水平明顯下降;與對照組比較,高脂高膽固醇組小鼠血清瘦素水平明顯升高[(7.14±2.44)ng/mL,](P0.01);與高脂高膽固醇組比較,1.5%、3.0%亮氨酸組小鼠瘦素水平[分別為(4.21±2.37)、(4.24±2.22)ng/mL]明顯降低(P0.01);與對照組比較,高脂高膽固醇組小鼠睪周脂肪組織中Ob-R蛋白表達下調;與高脂高膽固醇組比較,亮氨酸組小鼠睪周脂肪組織中Ob-R、JAK2蛋白表達上調、SOCS3蛋白表達下調。結論亮氨酸干預可抑制高脂高膽固醇喂養(yǎng)引起的小鼠體重和體脂增加,其機制可能與調節(jié)瘦素受體及其下游蛋白JAK2和SOCS3表達、改善瘦素抵抗有關。
[Abstract]:Objective to investigate the effect of leucine on body fat and leptin resistance in mice fed with high fat and high cholesterol.Methods 60 male C57BL/6J mice were randomly divided into control group, high cholesterol group, low dose leucine group and high dose leucine group for 24 weeks.The levels of serum cholesterol and leptin were determined by Western blot to detect the protein expression of leptin receptor Ob-Rn Janus protein tyrosine kinase 2JAK2, signal transduction and activator of transcription 3 STAT3 and cytokine signal transduction inhibitor 3SOCS3.Results the body weight, fat weight and serum cholesterol level of mice fed with high fat and high cholesterol were significantly increased, compared with those of high fat and high cholesterol group, the body weight, fat weight and serum cholesterol level of leucine group were significantly lower than those of control group.The expression of Ob-R protein was down-regulated in the peri-testis adipose tissue of mice with high lipid and high cholesterol, and the expression of Ob-R- ~ (-) JAK2 protein was down-regulated in the peri-testis adipose tissue of leucine group as compared with that in high-lipid and high-cholesterol group.Conclusion Leucine intervention can inhibit the increase of body weight and body fat induced by high fat and high cholesterol feeding in mice. The mechanism may be related to the regulation of the expression of leptin receptor and its downstream protein JAK2 and SOCS3 and the improvement of leptin resistance.
