野木瓜對(duì)鎘致SD大鼠亞慢性肝、腎損傷保護(hù)作用機(jī)制初探

【摘要】:目的:探討野木瓜(Stauntonia chinensi,SC)對(duì)鎘致SD大鼠亞慢性肝腎損傷的保護(hù)作用及其可能機(jī)制。方法:雄性SD大鼠50只,進(jìn)行體重分層,根據(jù)隨機(jī)數(shù)字法分為5組,每組10只。分組如下:空白對(duì)照組(C組)、單獨(dú)染鎘組(Cd組)兩組每天灌胃與體重相應(yīng)的生理鹽水,野木瓜低、中、高劑量組(SCL:0.5 g/kg;SCM:1g/kg;SCH:2g/kg)每天灌胃相應(yīng)劑量野木瓜懸濁液,除C組外,其余4組在飲水中添加Cd Cl2(50mg/L),飼養(yǎng)90天后處死大鼠。飼養(yǎng)結(jié)束后麻醉取血、肝、腎,-80℃保存。血生化檢測(cè)肝、腎功能相關(guān)指標(biāo);測(cè)定肝、腎MDA、SOD、GSH-Px含量;原子吸收光譜法檢測(cè)肝、腎組織及對(duì)應(yīng)組織超微結(jié)構(gòu)鎘含量;觀察肝、腎臟病理以及超微結(jié)構(gòu)改變;實(shí)時(shí)熒光PCR檢測(cè)肝、腎Mt-1、Mt-2 m RNA表達(dá)情況;Western blot檢測(cè)MT蛋白相對(duì)表達(dá)量。結(jié)果:染鎘之后,Cd組肝、腎臟器系數(shù)均高于C組(P0.05);SCH組肝、腎臟器系數(shù)均較Cd組低(P0.05),SCH組效果優(yōu)于SCL組(P0.05);與C組相比,Cd組ALT、AST、肌酐、尿酸大幅升高(P0.05),SCM、SCH兩組ALT、AST、肌酐、尿酸均有不同程度降低(P0.05);肝臟病理結(jié)果顯示:與C組相比,Cd組肝臟細(xì)胞發(fā)生腫脹,細(xì)胞核固縮,炎性細(xì)胞浸潤(rùn)和壞死,野木瓜干預(yù)能明顯減輕鎘誘發(fā)的炎癥反應(yīng),減少肝細(xì)胞壞死,改善肝臟損傷;腎臟病理結(jié)果發(fā)現(xiàn):與C組相比,Cd組腎小球萎縮,腎小管上皮細(xì)胞發(fā)生腫脹,腎小管管腔變小,腎小管上皮細(xì)胞出現(xiàn)缺失壞死,野木瓜干預(yù)明顯減輕腎損傷,改善腎小管上皮細(xì)胞腫脹。染鎘之后,Cd組肝超微結(jié)構(gòu)的損害主要集中在線粒體以及內(nèi)質(zhì)網(wǎng)上,表現(xiàn)為內(nèi)質(zhì)網(wǎng)水腫,線粒體嵴內(nèi)隙水腫擴(kuò)張;Cd組腎超微結(jié)構(gòu)顯示,腎小球局部可見(jiàn)足細(xì)胞次級(jí)突起分布不均勻,基膜厚度不均勻;野木瓜各干預(yù)組能觀察到鎘對(duì)肝、腎超微結(jié)構(gòu)的損害有所減輕;原子吸收結(jié)果顯示:與Cd組相比,SCM、SCH兩組中肝、腎、線粒體、微粒體鎘含量明顯減少(P0.05);相較于Cd組,野木瓜干預(yù)組線粒體、微粒體中鎘含量減少,并且野木瓜干預(yù)組細(xì)胞質(zhì)鎘含量高于單獨(dú)染鎘組(P0.05);Cd組肝臟、腎臟脂質(zhì)過(guò)氧化指標(biāo)MDA含量升高,SOD、GSH-Px活性明顯降低(P0.05),給予野木瓜干預(yù)后,SCM、SCH兩組MDA含量均降低(P0.05),SOD、GSH-Px均有不同程度升高(P0.05)。RT-PCR檢測(cè)相關(guān)基因發(fā)現(xiàn):與Cd組相比,野木瓜干預(yù)各組肝、腎Mt-1 m RNA、Mt-2 m RNA表達(dá)增高(P0.05),且SCM、SCH組增高更為明顯;肝、腎組織MT蛋白印記結(jié)果顯示:與Cd組相比,SCM、SCH組MT蛋白相對(duì)表達(dá)顯著升高(P0.05)。結(jié)論:野木瓜對(duì)鎘致SD大鼠肝腎損傷具有保護(hù)作用。
[Abstract]:Objective: to investigate the protective effect and possible mechanism of Stauntonia chinensis SC) on subchronic liver and kidney injury induced by cadmium in SD rats. Methods: 50 male SD rats were divided into 5 groups according to random number method. Ten rats in each group were divided into the following groups: blank control group (C group) and cadmium group (CD group) (two groups were given daily normal saline corresponding to body weight.) in low, middle and high dose groups, SCL: 0.5 g 路kg ~ (-1) / kg SCM: 1 g 路kg ~ (-1) 路kg ~ (-1) SCH: 2 g 路kg ~ (-1) / day, the corresponding dose of papaya suspension was given daily, except for C group.
