砷聯(lián)合MC染毒致小鼠肝損傷的效應研究

【摘要】:目的觀察砷(As)和微囊藻毒素(MC)單獨及聯(lián)合染毒對小鼠血清及肝組織肝功能指標和氧化損傷指標的影響。方法 40只健康KM小鼠隨機分為4組,分別為對照組、染砷組(12.5μg/L)、染MC組(0.25μg/kg·dl)和聯(lián)合染毒劑量組(12.5μg/L+0.25μg/kg·dl)。腹腔注射染毒10周,測定血清和肝組織中ALT、AST、MDA、SOD和GSH-Px水平。結果各染毒組血清白蛋白和ALT的水平與對照組比較差異均無統(tǒng)計學意義(P0.05);與對照組比較,各染毒組的AST水平均升高,其中染MC組和聯(lián)合染毒組差異有統(tǒng)計學意義(P0.05);與對照組相比,染砷組和聯(lián)合染毒組SOD和GSH-Px活性均增高,差異有統(tǒng)計學意義(P0.05)。MDA變化不明顯。與對照組比較,肝組織中的白蛋白降低,變化有統(tǒng)計學意義(P0.05);與對照組比較染MC組和聯(lián)合染毒組的ALT、AST水平均有升高,且差異有統(tǒng)計學意義(P0.05),各組間MDA含量差異無統(tǒng)計學意義(P0.05),各組間SOD活性呈下降趨勢,但差異無統(tǒng)計學意義(P0.05);GSH-Px活性在各染毒組均顯著增高,有統(tǒng)計學意義(P0.05)。肝組織DNA在染砷及染MC組時有損傷,聯(lián)合染毒時損傷更明顯。結論低劑量MC-LR及砷和MC聯(lián)合染毒可以誘導小鼠肝功能損傷和DNA損傷,但抗氧化酶在染毒組增高,可對抗其氧化損傷作用。
[Abstract]:Objective to observe the effects of arsenic As) and microcystin MCV on liver function and oxidative damage in serum and liver tissue of mice. Methods 40 healthy km mice were randomly divided into four groups, control group. The arsenic group (12.5 渭 g / L), the MC group (0.25 渭 g / kg 路dl) and the combined dose group (12.5 渭 g / L 0.25 渭 g / kg 路dll) were injected intraperitoneally for 10 weeks. Sod and GSH-Px levels in serum and liver tissue were measured. Results the levels of serum albumin and ALT in each exposed group were not significantly different from those in the control group (P 0.05), and the AST levels in each exposure group were higher than those in the control group. Compared with the control group, the activities of SOD and GSH-Px in the arsenic exposed group and the combined exposure group were increased, and the difference was not significant in the MC group and the combined exposure group, and the difference was not significant compared with the control group. Compared with the control group, the level of alt AST in MC group and combined group was increased. The difference was statistically significant (P 0.05). There was no significant difference in MDA content among the groups. The activity of SOD decreased in each group, but the activity of GSH-Px increased significantly in all groups. DNA in liver tissue was damaged in arsenic and MC groups, especially in combined exposure. Conclusion low dose MC-LR and combined arsenic and MC could induce liver function damage and DNA damage in mice, but antioxidant enzymes were increased in the exposed group. It can resist its oxidative damage.
