p,p’-DDE宮內(nèi)暴露致雄性子代大鼠的生殖毒性

【摘要】:[目的]探討宮內(nèi)暴露p,p’-DDE對(duì)雄性子代生殖毒性的影響。[方法]母鼠交配成功后,將其隨機(jī)分為對(duì)照(玉米油)組和100 mg/kg p,p’-DDE染毒組,每組8只。染毒組于孕第8~15 d采用灌胃方式給予100 mg/kg p,p’-DDE處理,對(duì)照組孕鼠給予等容積玉米油。孕鼠自由分娩,觀察仔鼠個(gè)數(shù)和出生性別,測(cè)量雄性仔鼠肛殖距及檢查乳頭存留情況;計(jì)算睪丸的臟器系數(shù);利用計(jì)算機(jī)輔助精子分析系統(tǒng)測(cè)定雄性仔鼠的精子質(zhì)量各相關(guān)指標(biāo);酶聯(lián)免疫吸附法測(cè)定雄性子代血清睪酮水平。[結(jié)果]染毒組仔鼠肛殖距[(0.94±0.12)cm]較對(duì)照組仔鼠肛殖距[(1.11±0.13)cm]有所縮短,乳頭存留率(34%)較對(duì)照組(0%)明顯上調(diào),精子數(shù)目[(50.00±4.62)×106個(gè)/m L],較對(duì)照組[(70.63±4.17)×106個(gè)/m L]明顯降低,精子活力[(62.42±3.32)%],較對(duì)照組[(76.75±2.68)%]明顯下降,差異均有統(tǒng)計(jì)學(xué)意義。2組雄性仔鼠雌雄比例、睪丸臟器系數(shù)、血清睪酮水平、睪丸重量差異均無統(tǒng)計(jì)學(xué)意義。[結(jié)論]宮內(nèi)暴露p,p’-DDE可誘導(dǎo)雄性仔鼠出現(xiàn)雌性化特征,精子數(shù)目和活力下降,導(dǎo)致雄性子代生殖毒性。
[Abstract]:[objective] to investigate the effect of intrauterine exposure of ppna-DDE on the reproductive toxicity of male offspring. [methods] after mating successfully, the female mice were randomly divided into control group (corn oil) group and 100 mg/kg pmegue-DDE exposure group. Eight rats in each group were treated by oral administration of 100 mg/kg mg/kg pPER-DDE on the 15th day of gestation, while the control group was given equal volume corn oil. The pregnant rats were given free delivery, and the number and sex of the newborn rats were observed. The anus distance and nipple retention were measured, the visceral coefficient of testis was calculated, the sperm quality of male offspring was measured by computer assisted sperm analysis system, and the relative indexes of spermatozoa quality were measured by computer assisted sperm analysis system. The serum testosterone level of male offspring was measured by Elisa. [results] the anal length [0.94 鹵0.12 cm] of the exposed group was shorter than that of the control group [1.11 鹵0.13 cm], and the nipple retention rate (34%) was significantly higher than that of the control group. The number of sperm [50.00 鹵4.62) 脳 106 / mL] was significantly lower than that of the control group [70.63 鹵4.17) 脳 106 / mL] and sperm motility [62.42 鹵3.32 渭 m], which was significantly lower than that of the control group (76.75 鹵2.68%). There were significant differences between male and female ratio, testicular organ coefficient and serum testosterone level. There was no significant difference in testicular weight. [conclusion] Intrauterine exposure of pPG-DDE could induce male offspring to appear feminization characteristics, sperm number and motility decreased, resulting in reproductive toxicity of male offspring.
