ELISA檢測(cè)小麥粉中脫氧雪腐鐮刀菌烯醇的優(yōu)化

【摘要】:目的針對(duì)ELISA法檢測(cè)脫氧雪腐鐮刀菌烯醇(DON),對(duì)小麥粉樣品中DON提取和DON標(biāo)準(zhǔn)工作液制備進(jìn)行優(yōu)化。方法以超聲提取法提取小麥粉樣品中的DON,以含10%甲醇的DON陰性樣品提取液作DON的ELISA標(biāo)準(zhǔn)工作液溶解相,對(duì)檢測(cè)方法進(jìn)行優(yōu)化,并以優(yōu)化的方法對(duì)50份小麥粉樣品進(jìn)行檢測(cè)。結(jié)果檢測(cè)方法經(jīng)優(yōu)化后,樣品經(jīng)超聲提取僅需10 min,2 500.00 ng/g、500.00 ng/g、100.00 ng/g 3個(gè)濃度水平的平均回收率為94.69%~102.71%,平均CV%為6.50%,50份小麥粉樣品DON檢出率為30.00%,DON污染水平在57.26~112.37μg/kg,中位數(shù)為84.12μg/kg,所有樣品中DON含量均未超過國家規(guī)定的限量標(biāo)準(zhǔn)。結(jié)論優(yōu)化后的檢測(cè)方法,大大縮短了樣品提取時(shí)間,提高了樣品提取率,降低了樣品基質(zhì)的干擾,方法重復(fù)性良好。
[Abstract]:Objective to detect the deoxynivalenoldol in wheat flour by ELISA, to optimize the extraction of DON from wheat flour and the preparation of standard working solution of DON. Methods the method of ultrasonic extraction was used to extract DON from wheat flour, and the negative sample of DON containing 10% methanol was used to extract DON from wheat flour. The product extract was used as the ELISA standard working liquid dissolution phase of DON. The detection method was optimized, and 50 wheat flour samples were detected by the optimized method. The average recovery of the three concentrations of 100.00 ng/g was 94.69 ~ 102.71, and the detection rate of DON in 50 wheat flour samples was 6.50%. The detection rate of DON was 30.002. 00 渭 g / kg at 57.26112.37 渭 g / kg, median 84.12 渭 g / kg, and the DON content in all samples was not higher than that in the country, and the average recovery was 94.69% 102.71%, and the average CV% was 6.50%.
