【摘要】：目的比較實驗室自制酶底物濾膜測試片法和51孔定量盤酶底物法在檢測水中總大腸菌群與大腸埃希菌上是否具有等效性。方法選取不同地區(qū)的3個實驗室,分別采集生活飲用水和水源水,同時采用酶底物濾膜測試片法和51孔定量盤酶底物法進(jìn)行總大腸菌群和大腸埃希菌檢測,按照ISO 17994∶2014《水質(zhì)-微生物方法之間的等效性建立標(biāo)準(zhǔn)》對檢測結(jié)果數(shù)據(jù)進(jìn)行分析。結(jié)果 3個實驗室采用兩種方法檢測總大腸菌群均數(shù)的擴展不確定度的置信上限范圍為(0.0494,0.0903),置信下限范圍為(-0.0713,-0.0490);檢測大腸埃希菌均數(shù)的擴展不確定度的置信上限范圍為(0.0307,0.0882),置信下限范圍為(-0.0928,-0.0381);兩者置信上限和下限均在可接受的區(qū)間范圍內(nèi)(0,0.10000)和(-0.1000,0)。說明3個實驗室采用兩種方法檢測總大腸菌群與大腸埃希菌的等效性驗證結(jié)果均無差異。結(jié)論酶底物濾膜測試片法和51孔定量盤酶底物法在檢測總大腸菌群與大腸埃希菌參數(shù)上具有等效性,進(jìn)行總大腸菌群與大腸埃希菌檢測時,酶底物濾膜測試片法可以作為51孔定量盤酶底物法的替代和補充。
[Abstract]:Objective to compare the equivalence of enzyme substrate filter membrane test method and 51 hole quantitative enzyme substrate method in the detection of total coliform bacteria and Escherichia coli in water. Methods three laboratories in different regions were selected. Drinking water and source water were collected, and total coliform bacteria and Escherichia coli were detected by enzyme substrate method and 51 hole quantitative enzyme substrate method. According to ISO 17994: 2014, "criteria for establishing equivalence between Water quality and microbial methods", the data of test results were analyzed. Results the confidence upper limit of extended uncertainty of testing the mean number of total coliform bacteria by two methods was used in three laboratories. The range of confidence limits is 0.0494-0.0903, the lower confidence limit is 0.0713- 0.04900.The confidence upper limit of the extended uncertainty for the detection of Escherichia coli is 0.0307 0.0882, and the lower confidence limit is 0.0928- 0.0381g; the confidence upper and lower limits are both within the acceptable range of 0.10000 and 0.10000.100.The results show that the confidence upper and lower limits are within the acceptable range. There was no difference in the results of the equivalence between the two methods for the detection of total coliform bacteria and Escherichia coli in three laboratories. Conclusion the method of enzyme substrate filter membrane test and 51 hole quantitative diskette substrate method are used to detect total coliform bacteria and Escherichia coli. The parameters of the bacteria are equivalent. In the detection of total coliform and Escherichia coli, the enzyme substrate filter membrane method can be used as the substitute and supplement of 51 hole quantitative enzyme substrate method.
