雙歧醋對高脂飲食大鼠預(yù)防性減肥和降脂的實(shí)驗(yàn)研究

【摘要】:目的: 研究雙歧醋對高脂飲食大鼠預(yù)防性減肥和降脂的作用,以及對機(jī)制的探討。 方法: 1.雙歧醋對高脂飲食大鼠預(yù)防性減肥的實(shí)驗(yàn)研究:將SD大鼠隨機(jī)分成雙歧醋低、中、高劑量組、市售醋組、肥胖模型組以及基礎(chǔ)對照組,灌胃法喂飼大鼠,除基礎(chǔ)對照組喂食基礎(chǔ)飼料外,其余組均喂食高營養(yǎng)飼料。測定指標(biāo)包括大鼠體重、體脂、Lee′s指數(shù)、脂肪細(xì)胞直徑、血瘦素、血胰島素。 2.雙歧醋對高脂飲食大鼠血脂的影響:將SD大鼠隨機(jī)分成雙歧醋低、中、高劑量組、市售醋組、高脂模型組以及基礎(chǔ)對照組,灌胃法喂飼大鼠,除基礎(chǔ)對照組喂食基礎(chǔ)飼料外,其余組均喂食高脂營養(yǎng)飼料。測定指標(biāo)包括大鼠體重、肝指數(shù)、體脂指數(shù)、病理學(xué)觀察肝臟脂肪變性。 3.雙歧醋對高脂飲食大鼠預(yù)防性降脂的機(jī)制探討:將SD大鼠隨機(jī)分成雙歧醋低、中、高劑量組、市售醋組、高脂模型組以及基礎(chǔ)對照組,灌胃法喂飼大鼠,除基礎(chǔ)對照組喂食基礎(chǔ)飼料外,其余組均喂食高脂營養(yǎng)飼料。測定指標(biāo)包括大鼠肝臟脂質(zhì)、肝臟脂肪變性、低密度脂蛋白受體(LDLR)的免疫組化分析、羥甲基戊二酸單酰CoA(HMGCoA)還原酶和LDLR的RT-PCR分析。 結(jié)果: 1.與肥胖模型組比較,雙歧醋各組大鼠的體重、體脂、Lee′s指數(shù)、脂肪細(xì)胞直徑、血瘦素、血胰島素均有明顯好轉(zhuǎn),特別是血瘦素和血胰島素水平。 2.與高脂模型組比較,雙歧醋各組的大鼠體重、肝指數(shù)、體脂指數(shù)、肝臟脂肪變性程度均有明顯好轉(zhuǎn),,特別是甘油三酯(TG)和高密度脂蛋白膽固醇(HDL-C)水平。 3.雙歧醋對HMGCoA還原酶表達(dá)有抑制作用,但可以上調(diào)LDLR的基因和蛋白質(zhì)表達(dá)。 結(jié)論: 雙歧醋在預(yù)防高脂飲食大鼠肥胖和血脂升高方面有一定的作用,并有劑量效應(yīng)關(guān)系。
[Abstract]:Objective: To study the effect of bifidobacterium vinegar on preventive weight loss and lipid lowering in rats with high fat diet and to explore the mechanism. Methods: 1. Experimental study of Bifidobacterium Acetate on Preventive weight loss in Rats with High Fat Diet: SD rats were randomly divided into three groups: low, medium, high dose, market vinegar, obesity model and basic control group. The rats were fed with the basic diet except the basic control group and the other groups were fed with the high nutrition diet. The parameters included body weight, body fat Leehs index, adipocyte diameter and blood leptin. Blood insulin. 2. Effects of Bifidobacterium Acetate on Blood Lipid in Rats with High Fat Diet: SD rats were randomly divided into three groups: low, medium, high dose, commercial vinegar, hyperlipidemia model and basic control group. All of the control groups were fed with high fat nutritional diet except the basic control group, including body weight, liver index, body fat index and pathological observation of hepatic steatosis. 3. Mechanism of Bifidobacterium Acetate on preventing Lipid lowering in Rats with High Fat Diet: SD rats were randomly divided into three groups: low, medium, high dose, market vinegar, hyperlipidemia model and basic control group. The rats were fed by gavage, except the basic diet for the basic control group, and the other groups were fed with the high fat nutritive diet, including the lipids of the liver and fatty degeneration of the liver. Immunohistochemical analysis of low density lipoprotein receptor (LDLR), RT-PCR analysis of hydroxymethyl glutaric acid monoyl CoA (HMG CoA) reductase and LDLR. Results: 1. Compared with the obese model group, the body weight, body fat Leehs index, adipocyte diameter, blood leptin and serum insulin of the rats in the bifidobacterium vinegar group were significantly improved. In particular, serum leptin and insulin levels. 2.Compared with the hyperlipidemia model group, the body weight, liver index, body fat index and degree of hepatic steatosis of the rats in the bifidobacterium vinegar group were obviously improved. In particular, the levels of triglyceride (TG) and high density lipoprotein cholesterol (HDL-C). 3. Bifid vinegar can inhibit the expression of HMGCoA reductase, but can up-regulate the expression of LDLR gene and protein. Conclusion: Bifidobacterium vinegar plays a certain role in preventing obesity and hyperlipidemia in rats with high fat diet and has a dose-effect relationship.
