1800MHz射頻電磁場(chǎng)通過瞬時(shí)受體電位C通道影響原代大鼠皮層神經(jīng)元突起生長

【摘要】:目的研究1 800 MHz射頻電磁場(chǎng)(radio frequency electromagnetic field,RF-EMF)對(duì)原代培養(yǎng)皮層神經(jīng)元突起生長的影響,并通過瞬時(shí)受體電位C通道介導(dǎo)的鈣內(nèi)流,探討RF-EMF對(duì)突起生長影響的機(jī)制。方法將原代培養(yǎng)皮層神經(jīng)元(primarily cultured cortical neurons,PCCNs)用隨機(jī)數(shù)表法分為4組:假暴露(Sham)、Sham+SKF-96365、輻照組(RF)和RF+SKF-96365組。使用s Xc-1800輻照系統(tǒng),1 800 MHz頻率talk模式,5 min開10 min關(guān),比吸收率為4 W/kg,輻照組輻照時(shí)間分別為12、24 h和48 h。SKF-96365干預(yù)處理是在維持培養(yǎng)基中加入SKF-96365(5μmol/L)后與假暴露組、RF組進(jìn)行相同的處理。輻照結(jié)束后即刻進(jìn)行以下指標(biāo)檢測(cè):1細(xì)胞活力(CCK-8法)和LDH釋放;2TRPC1、TRPC3蛋白表達(dá)(Western blot);3鈣庫操縱性鈣內(nèi)流變化(store-operated calcium entry,SOCE)(激光共聚焦顯微鏡);4神經(jīng)元突起總長度、初級(jí)突起數(shù)和分支數(shù)(Tuj-1免疫熒光染色)。結(jié)果 RF組細(xì)胞活力和LDH釋放與相應(yīng)Sham組相比差異均無統(tǒng)計(jì)學(xué)意義(P0.05)。與Sham組比較,RF組輻照24 h后,TRPC1和TRPC3蛋白表達(dá)分別降低24%和23%,SOCE下降54%,輻照48 h引起PCCNs突起總長度、初級(jí)突起數(shù)和分支數(shù)分別下降20%、12%、31%,以上變化均有統(tǒng)計(jì)學(xué)意義(P0.05)。Sham+SKF-96365組SOCE下降24%,突起總長度、初級(jí)突起數(shù)、分支數(shù)分別下降10%、12%、24%,與Sham組比較均有統(tǒng)計(jì)學(xué)意義(P0.05),而RF+SKF-96365組SOCE和突起總長度、突起數(shù)、分支數(shù)的變化與RF組比較差異無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論 RF-EMF可引起原代皮層神經(jīng)元突起長度、初級(jí)突起數(shù)和分支數(shù)減少,該效應(yīng)可能與TRPC1和TRPC3通道蛋白表達(dá)降低和TRPC通道介導(dǎo)的鈣信號(hào)減弱有關(guān)。
[Abstract]:Objective To investigate the effects of 1,800 MHz radio frequency electromagnetic field ( RF - EMF ) on the growth of neurons in primary cultured cortical neurons , and to investigate the mechanism of RF - EMF on the growth of neurons by transient receptor potential C - channel . Methods The primary cultured cortical neurons ( PCNs ) were randomly divided into four groups : sham exposure ( Sham ) , Sham + SKF - 96365 , irradiation group ( RF ) and RF + SKF - 96365 . Conclusion RF - EMF can decrease the protrusion length , the number of primary protrusions and the number of branches in the primary cortical neurons , which may be related to the decrease of TRPC and TRPC3 channel protein expression and the decrease of TRPC channel - mediated calcium signal .
