【摘要】:四棱豆是一種葉片、果實均可食用的新型糧、油、菜、藥兼用型的經(jīng)濟作物,目前已經(jīng)在我國南方推廣種植,本研究對四棱豆的功能成分及其特性進行了初步研究,為開發(fā)四棱豆?fàn)I養(yǎng)保健品提供理論基礎(chǔ)。 本研究利用原子熒光光譜法、紫外分光光度法、熒光分光光度計法對四棱豆的九個品種:湘棱豆一號、湘棱豆二號、湘棱豆三號、坪陽一號、中翼一號、Z4、 K00002、K000026、K000032的硒含量、維生素E含量和維生素C含量進行了檢測;采用(1,1-二苯基-2-三硝基苯肼)DPPH自由基清除法對九個品種的四棱豆抗氧化能力做出了評價,并對湘棱豆的提取方式進行了優(yōu)化選擇。結(jié)果表明,湘棱豆系列的Vc含量為13.09±0.31mg/100g, Ve含量為128.71+3.47mg/100g,硒含量為7.92±0.64μg/g。在保溫10min條件下的清除率為:湘棱豆一號為37.82%、湘棱豆二號為40.51%、湘棱豆三號35.67%。采用不同溶劑提取湘棱豆系列后,對DPPH自由基的清除效率為石油醚粗提物乙酸乙酯粗提物丙酮粗提物水粗提物乙醇粗提物。 建立低硒大鼠模型,采取灌胃的方式對大鼠補充高低兩個劑量組的亞硒酸鈉、富硒酵母和四棱豆硒蛋白溶液,研究不同硒源對大鼠生長發(fā)育、臟器系數(shù)、硒含量、抗氧化活力等生理指標的影響。將大鼠隨機分成6個處理組,每組8只,進行為期60d的生存試驗。試驗結(jié)果表明:各處理組大鼠與空白組大鼠的肝臟系數(shù)均有顯著性差異(P0.05),四棱豆硒蛋白高劑量組大鼠體重與空白組有顯著差異(P0.05);各處理組大鼠硒含量、SOD活性、MDA含量、GSH-px活性和TrxR活性均與對照組有顯著性差異(P0.05),其中亞硒酸鈉組大鼠組織硒含量與富硒酵母組和四棱豆硒蛋白組有顯著性差異(P0.05),四棱豆硒蛋白高劑量組的血清和肝臟SOD活力提升高達17.7%和21.2%,而四棱豆硒蛋白和富硒酵母兩個劑量組的GSH-px和TrxR活性均與亞硒酸鈉組有顯著差異(P0.05)。 以高血脂大鼠為試驗?zāi)P?采用湘棱豆乙醇提取物三個劑量組灌胃處理高血脂大鼠,并用辛伐他汀藥物作為陽性對照組,考察大鼠的生長情況、SOD酶活、MDA含量、血清轉(zhuǎn)氨酶活力和脂質(zhì)代謝情況。結(jié)果表明,湘棱豆提取液濃度為7mg/mL組抗氧化效果最明顯,體重較造模時有一定下降,WBEE(四棱豆乙醇提取物)高劑量組的腎周脂和睪丸周脂相對于高血脂模型組下降了22.22%和18.28%,肝臟重量也降低為12.76g,H-WBEE的肝臟勻漿MDA含量下降了13.43%,H-WBEE組谷丙轉(zhuǎn)氨酶和谷草轉(zhuǎn)氨酶分別下降了13.6%和11.7%,TG、 TC、LDL-c含量分別下降了36.2%、23、4%.66.7%, HDL-c含量增加了35.1%。 結(jié)果表明,湘棱豆品系的Vc、Ve、硒含量和自由基的清除率均高于其他品種,采用乙醇提取湘棱豆的粗提物對DPPH自由基的清除率高于其他提取劑。四棱豆硒蛋白組的抗氧化活性高于亞硒酸鈉組和富硒酵母組,而以乙醇提取后的WBEE高劑量組能明顯改善大鼠肥胖和降血脂,與降脂藥物效果相差不大。
[Abstract]:Tetragonia bean is a new kind of cash crop with edible fruit and edible grain, oil, vegetable and medicine. It has been popularized and planted in the south of China. In this study, the functional components and characteristics of tetragonia bean were studied preliminarily, which provided a theoretical basis for the development of nutrition and health products of quadrangular bean. In this study, atomic fluorescence spectrometry, ultraviolet spectrophotometry and fluorescence spectrophotometer were used to study nine varieties of tetragonia: Xianglingdou 1, Xianglingdou 2, Xiangleng bean 3, Pingyang 1 and Zhongyi 1. The selenium content, vitamin E content and vitamin C content of Z4, K00002, K000026 and K000032 were determined. The antioxidant ability of nine varieties of tetragonia bean was evaluated by DPPH free radical scavenging method of 1-diphenyl-2-trinitrophenylhydrazide. The results showed that the VC content of Xiangleng bean was 13.09 鹵0.31 mg / 100 g. The content of ve was 128.71 3.47 mg / 100 g. The selenium content was 7.92 鹵0.64 渭 g / g. The clearance rate of Xianglingdou 1 and Xianglingdou 2 was 37.82 and 40.51% respectively. The scavenging efficiency of DPPH free radical was: crude petroleum ether extract, ethyl acetate extract, acetone extract, ethanol crude extract. The rat model of low selenium was established, and sodium selenite, selenium enriched yeast and tetragonium bean selenium protein solution were added to the rats by gavage to study the growth and development of rats with different selenium sources and viscera coefficient. The effect of selenium content, antioxidant activity and other physiological indexes. The rats were randomly divided into 6 treatment groups, 8 rats in each group. The survival test was conducted for 60 days. The results showed that the liver coefficients of each treatment group and blank group were significantly different (P 0.05). There was significant difference in body weight between the high dose group and the blank group (P 0.05). Se content sod activity MDA content GSH-px activity and TrxR activity of rats in each treatment group were significantly different from those of the control group (P0.05). The content of selenium in sodium selenite group was significantly different from that in selenium enriched yeast group and tetrahedral bean protein group (P0.05). The activity of SOD in serum and liver was increased by 17.7% and 21.2% in the high dose group of selenoprotein. The activities of GSH-px and TrxR in the selenoprotein and selenium-enriched yeast groups were significantly different from those in the sodium selenite group (P 0.05). Taking hyperlipidemia rats as the experimental model, three dose groups of ethanol extract of Xiangleng bean were used to treat rats with hyperlipidemia, and Simvastatin was used as positive control group to investigate the growth of rats and the activity of SOD. The content of MDA, the activity of serum aminotransferase and the metabolism of lipid. The results showed that the antioxidation effect was the most obvious in the group of 7mg / mL extract of Xiangleng bean, and the body weight was lower than that of the model. Compared with the hyperlipidemia model group, the perirenal lipid and testicular pericarpium decreased by 22.22% and 18.28, and the liver weight decreased to 12.76g in the high dose group. The content of MDA in liver homogenate of H-WBEE decreased by 13.43% and decreased by 13.6% and 11.7T respectively in H-WBEE group. The content of LDL-c decreased 36.2%, and the content of HDL-c increased 35.1g. The results showed that the content of selenium and the scavenging rate of free radical in Xiangleng bean were higher than those in other varieties. The scavenging rate of DPPH free radical of the crude extract extracted by ethanol was higher than that of other extracts, and the antioxidant activity of selenoprotein group was higher than that of sodium selenite group and selenium-enriched yeast group. However, the high dose group of WBEE extracted with ethanol could obviously improve obesity and reduce blood lipid in rats, and the effect was not different from that of antilipidemic drugs.
