金剛戊脯腈對(duì)糖尿病神經(jīng)元損傷的保護(hù)作用及機(jī)制

目的:觀(guān)察DPP-Ⅳ抑制劑金剛戊脯腈(Adapentpronitrile,APPN)對(duì)糖尿病神經(jīng)元損傷的保護(hù)作用及機(jī)制。方法:1.APPN對(duì)糖尿病神經(jīng)元損傷的作用觀(guān)察:給予雄性SD大鼠(80-100g)高脂高糖飲食4周,腹腔注射鏈脲佐菌素(STZ 30mg/kg)并繼續(xù)高脂飲食4周,建立糖尿病大鼠模型。設(shè)立對(duì)照組(CMC-Na);糖尿病組(CMC-Na);糖尿病+高劑量組(APPN 4.5mg/kg);糖尿病+低劑量組(APPN 1.5mg/kg)。每組7只,連續(xù)灌胃給藥一個(gè)月,每天一次,自由飲食。用HE染色觀(guān)察大鼠腦組織形態(tài)、WB檢測(cè)腦組織APP與Aβ表達(dá)情況,以考察糖尿病神經(jīng)元損傷模型建立是否成功;生化酶學(xué)方法檢測(cè)大鼠腦組織中SOD活性及MDA含量;WB檢測(cè)腦組織GLP-1R,以及線(xiàn)粒體凋亡相關(guān)蛋白BAX、Bcl-2、cytochrome c、caspse-9及caspse-3蛋白表達(dá)情況等。2.APPN的血及腦組織濃度測(cè)定:選取4只SD大鼠(180-200g),禁食不禁水6h,尾靜脈注射APPN(50mg/kg)并于30min后腹主動(dòng)脈取血并分離腦組織,用HPLC-UV法檢測(cè)血漿...
