天冬氨酸對(duì)脂多糖誘導(dǎo)的仔豬肝臟損傷和肌肉蛋白質(zhì)降解的調(diào)控作用

【摘要】:本文研究了天冬氨酸(ASP)對(duì)脂多糖(LPS)刺激仔豬肝臟損傷和肌肉蛋白質(zhì)降解的調(diào)控作用及其機(jī)制。 1、本試驗(yàn)研究了ASP對(duì)LPS刺激仔豬肝臟損傷的保護(hù)作用,并從Toll樣受體4(TLR4)和核苷酸結(jié)合寡聚域受體(NOD)信號(hào)通路的角度探討其機(jī)制。選用48頭斷奶仔豬,分為4個(gè)處理組:(1)對(duì)照組(基礎(chǔ)日糧);(2)LPS組(基礎(chǔ)日糧+LPS);(3)0.5%ASP組(基礎(chǔ)日糧+0.5%ASP+LPS);(4)1.0%ASP組(基礎(chǔ)日糧+1.0%ASP+LPS)。試驗(yàn)期為21天。第20天,給2、3和4組注射100μg/kg體重的LPS,1組注射等量的生理鹽水,,4h和24h后采血。采血后屠宰,取肝臟樣品待測(cè)。結(jié)果表明:(1)0.5%和1.0%ASP可緩解注射LPS4h和24h后肝臟形態(tài)結(jié)構(gòu)的損傷。(2)1.0%ASP可緩解注射LPS4h后血清堿性磷酸酶(AKP)和γ-谷氨酸轉(zhuǎn)肽酶活性升高;0.5%或1.0%ASP可緩解注射LPS24h后血清谷草轉(zhuǎn)氨酶、AKP活性的升高和谷丙轉(zhuǎn)氨酶/谷草轉(zhuǎn)氨酶比例的降低(P0.05)。(3)1.0%ASP可緩解注射LPS24h后claudin-1蛋白表達(dá)量的降低(P0.05)。(4)0.5%和1.0%ASP緩解了注射LPS4h后腫瘤壞死因子-α(TNF-α),環(huán)氧合酶2(COX2)和熱休克蛋白70mRNA表達(dá)量的升高,1.0%ASP緩解了注射LPS24h后TNF-α和COX2mRNA表達(dá)量的降低(P0.05)。(5)0.5%或1.0%ASP緩解了注射LPS4h后TLR4和其下游信號(hào)分子髓樣分化因子88、白介素受體相關(guān)激酶1和腫瘤壞死因子受體相關(guān)因子6,以及NOD1、NOD2和其下游信號(hào)分子受體互作蛋白激酶2mRNA表達(dá)量的上升,降低了核因子(NF)-κBp65的mRNA表達(dá)量(P0.05);0.5%或1.0%ASP緩解了注射LPS24h后TLR4,NOD1和NOD2的mRNA表達(dá)量的降低(P0.05)。這些結(jié)果表明:在LPS刺激早期和晚期,ASP對(duì)TLR4和NOD信號(hào)通路相關(guān)基因的mRNA表達(dá)、炎性介質(zhì)的產(chǎn)生發(fā)揮相反的調(diào)控作用,并改善了肝臟結(jié)構(gòu)和功能。 2、本試驗(yàn)研究了ASP對(duì)LPS刺激仔豬肌肉蛋白質(zhì)降解的保護(hù)作用,并從AMP激活蛋白激酶(AMPK)、蛋白激酶B/叉頭轉(zhuǎn)錄因子(Akt/FOXO)、TLR4和NOD信號(hào)通路的角度探討其機(jī)制。選用24頭斷奶仔豬,分為4個(gè)處理組:(1)對(duì)照組(基礎(chǔ)日糧);(2)LPS組(基礎(chǔ)日糧+LPS);(3)0.5%ASP組(基礎(chǔ)日糧+0.5%ASP+LPS);(4)1.0%ASP組(基礎(chǔ)日糧+1.0%ASP+LPS)。試驗(yàn)期為19天。第19天,給2、3和4注射100μg/kg體重的LPS,1組注射等量的生理鹽水,4h后屠宰,取肌肉樣品待測(cè)。結(jié)果表明:(1)0.5%ASP提高了腓腸肌DNA和RNA含量(P0.05);1.0%ASP提高了腓腸肌蛋白質(zhì)和RNA含量,提高了背最長(zhǎng)肌RNA/DNA比值和蛋白質(zhì)/DNA比值(P0.05)。(2)1.0%ASP緩解了注射LPS后腓腸肌肌萎縮F-box(MAFbx)和肌肉環(huán)指蛋白1(MuRF1)mRNA的表達(dá)量上升(P0.05),0.5%和1.0%ASP緩解了注射LPS后背最長(zhǎng)肌MuRF1mRNA的表達(dá)量上升(P0.05)。(3)0.5%或1.0%ASP降低了腓腸肌pAMPKα/tAMPKα,緩解了注射LPS后肌肉pAkt/tAkt和pFOXO1/tFOXO1降低,降低了背最長(zhǎng)肌FOXO4的mRNA表達(dá)量(P0.05)。(4)ASP沒(méi)有抑制肌肉TLR4信號(hào)通路相關(guān)基因的表達(dá),相反還提高了背最長(zhǎng)肌簇分化抗原14、MD2和脂多糖結(jié)合蛋白的mRNA表達(dá)量(P0.05)。(6)1.0%ASP提高了腓腸肌的細(xì)胞因子信號(hào)抑制物1(SOCS1)和toll反應(yīng)蛋白(Tollip),背最長(zhǎng)肌的RP105、SOCS1、Tollip和單免疫球蛋白IL-1受體相關(guān)分子的mRNA表達(dá)量(P0.05)。這些結(jié)果表明:ASP不是通過(guò)TLR4和NOD信號(hào)通路,而是通過(guò)抑制AMPKα磷酸化,提高Akt磷酸化,進(jìn)而提高FOXO磷酸化,從而抑制MAFbx和MuRF1表達(dá)來(lái)緩解肌肉蛋白質(zhì)降解。
[Abstract]:In this paper , the regulation and mechanism of lipopolysaccharide ( ASP ) on liver injury and muscle protein degradation in piglets stimulated by lipopolysaccharide ( LPS ) were studied .
1 . The protective effect of ASP on LPS - stimulated liver injury in piglets was studied , and its mechanism was discussed from the angle of Toll - like receptor 4 and nucleotide binding oligopeptide signal pathway . 48 weaned piglets were divided into 4 treatment groups : ( 1 ) control group ( basal diet ) ;
( 2 ) LPS group ( basal diet + LPS ) ;
( 3 ) 0.5 % ASP group ( basal diet + 0.5 % ASP + LPS ) ;
( 4 ) 1.0 % ASP group ( basal diet + 1.0 % ASP + LPS ) .
( 3 ) The levels of TNF - 偽 , COX2 and HSP70 mRNA were decreased after LPS24 injection ( P0.05 ) . ( 5 ) 0.5 % or 1.0 % ASP alleviated the increase of TNF - 偽 and COX2 mRNA expression after injection of LPS24 .
The results showed that after LPS stimulated early and late LPS stimulation , ASP had the opposite effect on the mRNA expression and inflammatory mediators , and improved the structure and function of liver .
2 . This study investigated the protective effects of ASP on muscle protein degradation in piglets stimulated by LPS , and discussed its mechanism from the angle of AMP - activated protein kinase ( AMPK ) , protein kinase B / protein kinase B / protein kinase B / protein kinase B / protein kinase B / protein kinase B / FOXO . The 24 - head weaned piglets were divided into 4 treatment groups : ( 1 ) control group ( basal diet ) ;
( 2 ) LPS group ( basal diet + LPS ) ;
( 3 ) 0.5 % ASP group ( basal diet + 0.5 % ASP + LPS ) ;
( 4 ) 1.0 % ASP group ( basal diet + 1.0 % ASP + LPS ) . The test period was 19 days . On day 19 , 100 渭g / kg body weight of LPS was injected into 2 , 3 and 4 groups of LPS , 1 group was injected with equal amount of physiological saline . After 4 hours , the muscle samples were slaughtered . The results showed that : ( 1 ) 0.5 % ASP increased the DNA and RNA content of calf muscle ( P0.05 ) ;
( 2 ) 1 . 0 % ASP reduced the mRNA expression level of the muscle atrophy F - box and the muscle ring finger protein 1 ( P0.05 ) .
